Boron is one of essential trace minerals element in plant growth and development. Howerer plants had a narrow range of concentration of boron, so boron concentration of the soil is too low will cause premature aging while too high can cause blade early fall phenomenon. Boron deficiency phenomenon is widespread in the citrus regions, and add boron fertilizer improper will cause toxicity seriously phenomenon, of which both of them will be affected the growth and development of citrus. Recent studies have shown that a special type of water channel protein, aquaporins MIP family proteins, were involved in the process of absorption and transportation of boron in plants; so the analysis of the transcription of citrus MIP gene family under the condition of boron adversity will help to understand the molecular mechanism of the citrus adapt to boron adversity.This study use Poncirus trifoliate which is the most common rootstock of citrus as the material and get the length of eighteen members of MIP family genes by cloning sequence, including ten PIPs and eight TIPs menbers, perfectly. The information of results is as follows:1、Bioinformatics analysis of Ptr PIPs and Ptr TIPs members(A) The results show that the length of Ptr PIPs and Ptr TIPs by clonging were the range of 750 bp to 1500 bp; of which most members were around 1000 bp.(B) Its two subfamily members and the five subfamily family of other species are located in different five branches, witch each other is homology in between, and had high homology with one’s in other species in phylogenetic tree; the Ptr PIPs subgroup was divided into two different subgroup, located at the next level branches.(C)Alignmented sequence and showed that these gene family had unique NPA(Asn-Pro-Ala) characteristic motifs, and had a characteristic signal sequence(-SGGHINPAVTFG).(D) The conservatived amino acid analysis showed that: The two subfamily members have their own unique conserved amino acid sites.(E) Analysised the tissue specificity and revealed that : Pr PIPs and Pr TIPs are mostly expressed in roots, the expression of part of members is in stems and leaves,and a few members of genens expressed in all different tissues; the other of which is stressed Ptr TIP5;1 mainly expression in stems, while both Ptr PIP2; 4 and Ptr PIP2; 7 expression mainly in leaves.2 、 The expression pattern of space and time of Ptr PIPs and Ptr TIPs members under boron stress(A) Under boron stress, the gene family members which responed to stress main were Ptr TIPs in roots, and Ptr PIPs in leaves, while Ptr PIPs and Ptr TIPs in stems.(B)The number of members which its expression were up regulated were seven in roots under different boron stress while down regulated were six. Among of them, the genes which the response to the tress intensely in roots were Ptr TIP4;1, Ptr TIP5;1 and Ptr PIP2;7 in turn under boron deficiency treatment. While the genes were Ptr TIP4;1,Ptr PIP2;7 and Ptr TIP1;3 under boron toxicity treatment.(C) The number of members which its expression were up regulated were eight in stems under different boron stress while down regulated were ten. Among of them, the genes which the response to the tress intensely in roots were Ptr TIP4;1, Ptr PIP2;7 and Ptr TIP1;3 in turn under boron deficiency treatment. While the genes were Ptr PIP2;7, Ptr TIP4;1and Ptr TIP1;3 under boron toxicity treatment.(D) The number of members which its expression were up regulated were ten in leaves under different boron stress while down regulated were six. Among of them, the genes which the response to the tress intensely in roots were Ptr PIP2;6, Ptr PIP2;4, Ptr TIP1;3 and Ptr TIP5;1 in turn under boron deficiency treatment. While the genes were Ptr PIP2;6, Ptr PIP2;4, Ptr TIP1;3and Ptr PIP2;2 under boron toxicity treatment.(E) Both in boron deficiency and boron toxicity treatment, Ptr PIP1;1 、 Ptr PIP1;2 、 Ptr PIP2;1 and Ptr TIP3;2 were down regulated in roots, stems and leaves,while Ptr PIP2;7and Ptr TIP4;1 were up regulated.
