Abstract/Summary:
615 strains isolated from different marinie environments were screened for the antimicrobial avtivity.2 strains of Bacillus with pesticide potential were identified. The growth characteristics and relevant control genes of 2 strains were determined.The important biocontrol genes were amplifled; The control effect of tomato leaf mould and rape sclerotinia for the strains were investigated. All the experimental results as follows:(1) Gene fragments related with antifungi activity of ituC, fenD, srfAA, srfAB, bioA and bmyB were found in 2 strains. The minimum inhibitory concentration (MIC) of 2 strains to A.solani were 1 μl/ml (C12-6) and 2μl/ml (Z10-3)and antimicrobial spectrum on Botrytis cinerea、Alternaria solani、Fusarium graminearum, Cladosporium fulvum、Sclerotinia sclerotirum、Fusarium oxysporum、Phytophthora capsici、Pyricularia grisea of the Z10-3strain from 74.1% to 85.4%, the C12-6 strain was from 63.1% to 83%. The control effects with the strain Z10-3 of tomato leaf mould and rape sclerotinia were 62.1%~63.6%, and the strain C12-6 were 59.1%~ 65.4%.(2) Gene clusters related with growth promoting were detected in 2 strains.Gene fragments related with indole acetic acids(IAA) of ysnE and yhcX were assayed in 2 strains and phy gene of phytase was found in Z10-3 strain.In 2 strains, gene fragments of alsS, alsD and alsR were found, which controlled synthesis of2,3-butanedio. The functions of 2 strains with phosphorus solubilization, potassium release, nitrogen fixation and IAA production were tested. C12-6 and Z10-3 strains have dissolved nitrogen fixation and phosphorus activity, and Z10-3 strain has the activity of releasing. C12-6 and Z10-3 strains can produce IAA and siderophore-producing activity were 28.9% and 68.7%.The results showed that 2 strains could colonize in rhizosphere soil, roots, stems and leaves of the tomato. The Z10-3 root colonization in simulate saline soil was preferable with the maximum amount of colonization were 60×106cfu/gand decreased 18×106cfu/g 15days later; The C12-6 root colonization in simulate saline soil was preferable with the maximum amount of colonization were 60×106cfu/g and decreased 15×106cfu/g 15days later. As for tomatos treated with 2 strains,the effect of tomato growth-promoting were significant by 2 strains, improved tomato fresh weight, plant height and root length., its chlorophyll content incressed 53.4%(C12-6r) and 57.4%(Z10-3r) at the Salt stress.(4) The Z10-3 strain was identified as Bacillus amyloliquefaciens by 16SrDNA, gyrB sequence, physiological and biochemical; The C12-6 was identified as Bacillus sp.
