Background:The heat stress is one of the factor that make oxidative stress produced. The special structure of polyphenols have a strong ability to capture oxygen free radicals, and it has a ability of antioxidant and scavenging radical. The laboratory has found that polyphenolic extract from involucres of Castanea mollissima Blume (PICB) have the ability of antioxidant and scavenging radical. And the study shown that the PICB can make the IEC-6 cells mRNA expression of antioxidant and growth factor up-regulation.Objective:The research explore the effect of PICB promote growth and antioxidation in IEC-6 cells, and the serum and jejunum mucosa metabolomics in AA broilers under heat stress. To explain the mechanism of PICB promote growth and antioxidation and then lay a foundation for PICB as a natural plant feed additive.Methods:1、Used the MTT to detected IEC-6 cells proliferation activity in different concentration of PICB impact and ascertain the added content.2. This test set for normal group (Ⅰ is control group and Ⅱ is PICB group.)and high group(Ⅲis control group and Ⅳis PICB group).Used the real-time PCR and Western Blotting to detected the impact of PICB to IEC-6 cells mRNA expression and protein expression.3, Used gas chromatography-time of flight-mass chromatography (GC-TOF-MS)to analysis the PICB to small molecule metabolites difference in serum and jejunum mucosa of AA broilers.Result:1、Under normal temperature, all concentration of PICB to IEC-6 cells have no significant difference compare with control group(P<0.05).Under heat stress, the control group have significant lower than PICB group and when the concentration at 180μg/ml, the OD value reached maximum (P<0.05).2、Under normal temperature, the group Ⅰ and group ⅡHSP70 and EGF mRNA expression have no significant change(P>0.05); group Ⅱ ODC、SOD and EGFR mRNA expression were higher than group I (P<0.05).the group Ⅱ protein expression of SOD is significant higher than group I (P<0.05),EGFR and ODC in group I and group Ⅱ have the same expression. Under heat stress, the mRNA expression of HSP70、ODC、EGF、EGFR、SOD in group Ⅳwere significant higher than group Ⅲ(P<0.05).the protein expression of SOD、ODC、EGFR in groupIVwere significant higher than group Ⅲ(P<0.05).4、Compared with the control group the 0.3% PICB group have significantly reduced the content of glutamate (P<0.05)in serum of AA broiler and reduce the content of inositol and threitol in jejunum mucosa(P<0.05), and significant improve the content of ornithine, serne and palmitic acid.Conclusion:PICB can significant improve the ability of growth and antioxidant under heat stress. in IEC-6 cells.That is to say PICB can relieve the heat damage in IEC-6 cells.0.3%PICB can improve the AA broilers growth performance and antioxidant to relieve the heat damage in AA broilers by glutathione, carbohydrate, lipid and protein metabolic pathway.
