Puccinia striiformis f. sp. Tritici(Pst) is the pathogen agent of the stripe trust, which causes damaging diseases of wheat. Recently, it has been discovered that basidiospore produced by teliospore can infect and complete the sexual stage on the berberis species. Though the morphology of teliospore’s formation has been reported, the infection biology of basidiospore is poorly understood. It’s widely known that urediospore infects wheat and causes stripe rust, but there are little reports about the interaction between urediospore and the alternate host of pathogen. So In our study, nuclear behavior during basidiospore formation, the interaction between basidiospore of Pst to the wheat as well as urediospore of Pst to the alternate host berberis species are investigated systematically through light microscopy and electron microscopy, which will lay the theatrical foundation for fully understanding of to basidiospore and urediospore infection biology. The fluorescence microscopy observation indicates that diploid teliospore undergoes a meiosis during formation of four haploid basidiospores, while basidiospore undergoes a mitosis and forms two nuclei before germination.The basidiospore can infect berberis in vitro successfully. The light microscope and electron microscope observations show that basidiospore can germinate and form germ tube normally on wheat the suspected papilla is believed to arrest the successive infection of pathogen. While, urediospore of Pst is able to invade the alternate host and form the haustorial mother cells. Our study provides the scientific basis to the deep analysis of Pst’s developmental biology.
