With the development of economy in recent years, people’s requirements and the quality for life are improving in all aspects, and it’s first reflected on the diet. China is a large agricultural country, animal husbandry is one of the important part, however, the traditional diet containing high greasy, caloric food has a great influence on our health. The rabbit meat has advantages such as low fat, low cholesterol and low calorie. And it can satisfy people’s requirements for healthy diet. In addition, our country has a big population and little land which resulted in the contradiction between grain supply and demand. Moreover, pigs and poultry are grain consuming livestock competing for food with people, which has become an increasingly prominent problem. Because the rabbit can effectively make use of the forage resources and it can save food, so it belongs to grain saving livestock. Therefore, it is great important for developing rabbitry to promote the economic development of China.However, the male rabbits have the sterile phenomenon in summer. When the rabbits are under the heat stress, it would lead to the decreased of semen volume, density, quality and so on. In order to solve the problem of summer breeding, we need to cryopreserve rabbit semen. However, the results of cryopreservation of rabbits semen is not satisfactory, therefore, it has not been widely used in the production practice.In order to further improve the quality of rabbit semen after frozen and thawed, our experiment was carried out to improve the antioxidant of dilution, by adding water-soluble vitamin E analogues(Trolox) to the cryopreservation solution. And we studied its effect on the quality of rabbit semen after cryopreservation. The evaluation index of frozen and thawed sperm contains the motility, membrane integrity, acrosome integrity, DNA integrity, lipid peroxidation level and mitochondrial activity. And we used propidium iodide(PI), SYBR-14, fluorescein isothiocyanate labeled with peanut agglutinin(FITC-PNA), acridine orange(AO), the fluorescence probe of BODIPY-C11 and 5,5’,6,6’-1,1’,3,3’-tetrachloro-tetraethylbenzim-idazolcarbocyanine iodide(JC-l) to assess the quality of sperm.The results are shown as follows:1.Rabbit semen cryopreservation with Trolox, after thawing each group of sperm motility, acrosome integrity and mitochondrial activity was significantly higher than the control group(P < 0.05), the lipid peroxidation levels were significantly lower than the control group(P < 0.05), when the concentration is 200μM, the plasma membrane integrity was significantly higher than the control group(P < 0.05).2. Comparison the effect of different concentration of Trolox on semen quality after thawing, on the concentration of 200μM, the sperm motility, acrosome integrity, mitochondrial activity was significantly higher than the concentration of 100μM and 250μM(P < 0.05), when the concentration is 100μM and 200μM, the lipid peroxidation level was significantly lower than the 250μM(P < 0.05).The results of this experiment show that vitamin E analogues can improve the freezing resistance ability of rabbit sperm and the optimum concentration of vitamin E analogues is 200μM.
