Abstract/Summary:
Neurocysticercosis is an important zoonotic parasitic disease, caused by Cysticercuscellulosae, the larva of Taenia solium, which parasitic on the central nervous system of thehost. And it is widely distributed in Latin America, Asia and Africa. In recent years, theprevalent tendency of the disease in American and European countries is also severe, and itattributed to the increased international trade, travel and migration. The disease not onlycaused a huge economic losses and endangering the public health. Thioredoxinperoxidase(TPx), also known as peroxiredoxin(Prx), are a class of low molecular weightproteins, is also an antioxidant in vivo to thioredoxin as the only hydrogen donor, the removalof the reactive oxygen species(ROS). In addition to the peroxide released TPx toxic effects,but also as a molecular chaperone, regulation of cell signal transduction mediated peroxide.Taenia solium Thioredoxin peroxidase (TsTPx) genes was obtained by RT-PCR with thetotal RNA of Taenia solium as template, bioinformatics analysis showed that the full-length ofthe gene mRNA was588bp, encoding195amino acids, the theoretical molecular weightproteins of21.59kDa.Protein TsTPx prokaryotically expressed in this study predominantly located in thesupernatant as a soluble protein confirmed by SDS-PAGE. Its antioxidative property wasproved by, positive correlation of TsTPx concentration to protection percentage of plasmid’ssuper-coiled DNA structure in metal catalytic oxidation system, where20μg/ml TsTPxachieved100%protection confirmed by disappearance of nicking slower band in agarosegelelectrophoresis; and also positive correlation of TsTPx concentration to H2O2clearancerate in H2O2reduction system, where100μg/ml TsTPx achieved respectively69.9%and54.5%of H2O2clearance with and without dithiothreitol. Its antigenicity was demonstrated byspecial recognition between Cysticercus positive serum from pig in Western blotting andantibody titers up to1:320000detected with indirect ELISA after TsTPx vaccine wasprepared and injected into rabbits.
