Effects Of The Zinc-enriched Yeast Fermentation Broth On Physiological Functions Of Digestive Tract In Model Mice

Zinc-enriched yeast is a new yeast strain by artificial mutation. Zinc-enriched yeastfermentation broth contains nutrition metabolites, flavor enhancers enzymes and other unknowngrowth factors. These elements can improve the diet palatability and animal digestibility. In thisstudy, the effects of the zinc-enriched yeast fermentation broth were investigated onphysiological functions of digestive tract of model mice. The mice were randomly divided into7groups: Ⅰ: the normal control group, Ⅱ: the CTX group, Ⅲ: the CTX+30%, Ⅳ: CTX+20%, Ⅴ:CTX+10%zinc-enriched yeast fermentation broth group, Ⅵ: the CTX+20%yeast fermentationbroth group and Ⅶ: the CTX+medium group. The dosage of gavage was determined byweighting. The immune injury mice model was induced by an intraperitoneal injection ofCytoxan(80mg/kg) on the11th day,continued for3days and killed on the15th day. The blood,gastric jejunum and liver of mice were collected for IgA, IgG, IgM, disaccharidase, pepsin,trypsin, LZM, ADA and Caspase-3detection. The liver and jejunum mucosa tissues wereobserved. Results indicated that:(1) Histologic changes Liver tissue structure: the hepaticsinusoid constriction, hepatic cell edema and vacuolated degeneration around the central veiwere found in the liver tissue of CTX group, hepatic cell were arranged mussily. The situationsof Ⅲ group were improved insignificantly, but Ⅳ, Ⅴ group and Ⅵ group significantly. Jejunumtissue structure: intestinal villi of the CTX group were shorten and loose. Part of epithelium wasfallen away. Mucosal epithelium were separated from lamina propria in the free-end of intestinalvilli. Intestinal mucosa were damaged, the intestinal villi were short and broken partly. Thesituations of zinc-enriched yeast fermentation broth groups were improved. The situation of Ⅴgroup were better than Ⅲ and Ⅳ groups. The Ⅵ and Ⅶ groups were not as good as Ⅴ group.(2)Immunoglobulins The content of IgA in serum tended to be increased by zinc-enriched yeastfermentation broth and were significantly increased in all groups (P<0.05) except Ⅲ group(P>0.05) compared with CTX group, especially, the Ⅴ group were highest; compared with CTXgroup, the content of IgG in serum was reduced by30%zinc-enriched yeast fermentation broth,Ⅳ and Ⅴ groups was increased (P>0.05); compared with CTX group, the content of IgM inserum were significantly increased in Ⅳ, Ⅵ and Ⅶ group (P<0.05), other groups had nosignificant difference(P>0.05).(3) Digestive enzymes activities Compared with nature group, the maltase and invertase activities of CTX group were reduced(P>0.05), but lactase activities ofCTX group increased(P>0.05); compared with nature group and CTX group, the maltaseactivities of Zinc-enriched yeast fermentation broth groups were significantly reduced (P<0.05),especially the Ⅳ group were lowest. The invertase and lactase activities of Zinc-enriched yeastfermentation broth groups were significantly increased(P<0.05), especially the Ⅲ group werehighest. Compared with the CTX group, the pepsin activities of Ⅲ group were significantlyreduced (P<0.05) and the trypsin activities were significantly increased (P<0.05). The trypsinactivities and pepsin activities of Ⅳand Ⅴ groups tended to fall (P>0.05).(4) Lysozymeactivities Compared with the CTX group, the lysozyme activities of zinc-enriched yeastfermentation broth groups were reduced, especially the Ⅲ group were lowest, and the threegroups had no significant difference(P>0.05). Compared with the normal control group and CTXgroup, the lysozyme activities of Ⅵ group and Ⅶ group had no significant difference comparedwith the CTX group and normal group(P>0.05).(5) ADA activities Compared with the CTXgroup, ADA activities of Ⅲ group had no significant difference(P>0.05), but ADA activities ofother groups were significantly reduced(P<0.05) and had no significant difference among thethree groups (P>0.05).(6) Caspase-3activities The activities of Caspase-3of experimentalgroups were significantly reduced (P<0.05) compared with the normal control group and had nosignificant difference compared with the CTX group (P>0.05). The activities of Caspase-3hadno significant difference among zinc-enriched yeast fermentation broth groups.It shows that Zinc-enriched yeast fermentation broth can protect liver tissue and jejunumtissue structure, and improve the situation of immune injury tissue. It can increase the contents ofIgA、IgG and IgM and enhance the immune responses abilities, the contents of IgA and IgG ofⅤ group are highest and the contents of IgM of Ⅳ group are highest. It can regulate digestiveenzyme activities of the digestive tract in injury mice induced by CTX: inhibit maltase, pepsinand trypsin activities and improve invertase and lactase activities, antagonize the damage ofgastric mucosa. It can reduce the contents of lysozyme and regulate micro-ecologicalenvironment steady state of digestive tract. It can reduce the contents of ADA, improve andrecover the structure and digestion function disorder in mice digestive tract, and the Ⅴ group arelowest. It also can reduce Caspase-3activities, extend the cells themselves apoptosis metabolismand maintain the mucosal cells’ normal metabolism cycle, it may contains some composition similarly to the substance in body related to digestion, so it can reduce the biosynthesis of theserelated substance in body.
