Study On Herbicide-resistant Soybean Transformation

Abstract/Summary:
Agrobacterium-mediated soybean transformation of cotyledonary-node was acommon-used method in soybean genetic transformation. The efficiency of thesystem was affected by all factors involved in genotype, germinating condition, shootinduction and rooting. We chose Dongnong50from11tested cultivars as the bestreceptor to establish Agrobacterium-mediated soybean transformation system ofcotyledonary-node. The main results were as below:The induction rate was72.34%, and the highest elongation rate was20.59%. Inrooting, dipping the stem in1mg·L-1IBA for1minute could be helpful for thisprocess and the rooting rate was85.25%.10of18T0plants was positive of both barand EPSPS gene. PCR analysis showed that the efficiency of this transformationsystem achieved7.05%.We established the criteria system of phosphinothricin-resistance. The painting concentrationof phosphinothricin was100mg·L-1.1PCR positive plants of T0had phosphinothricin-resistant of100mg·L-1. In T1generation,4plants had phosphinothricin-resistance of100mg·L-1,6hadphosphinothricin-resistance of150mg·L-1and2had phosphinothricin-resistance of200mg·L-1.
