Triticale could be used to produce grain and high quality forages to provide the concentrateand feed for the development of livestock industry. In this study,59triticale lines were chosen asthe experimental materials, their genetic diversity, productivity and quality in the couty of Lintaoand Maqu, Gansu province were studied. Based on this study, we could know the geneticrelationship of these germplasm and determine the suitable sowing date and area. Main resultswere as follows,Using L16(45) orthogonal design, the optimization of the triticale ISSR-PCR optimalreaction system were as follows, dNTPs0.4mmol/L,Mg2+2mmol/L,Primers0.5μmol/L,Taqpolymerase0.5U. PCR amplification procedure were: pre modification at94oC for7min;modification at94oC for30s, annealing for45s,72oC for1min,35cycles; at last, extension for7min at72oC, and saved at4oC for use.According to cluster analysis,59triticale strains were divided into8groups while thegenetic similarity coefficient was0.76. Lines J2, J7, J12, J14, J15, J30and J51were classified asthe type II; J10and J24were classified as the type III; J28belonged to the type IV; J11, J26, J27,J37, J45, J46, J47, J57and J58belonged to the type V; J5, J17, J23and J38belonged to the typeVI; J16and J19belonged to the type VII; J18, J20, J21and J22are the type of VIII; and theothers were classified as the type I.Seed yields for lines J1, J3, J5, J8, J10, J11, J12, J14, J15, J16, J17, J18, J23, J24, J28, J30, J32,J33, J34, J35, J37, J38, J40, J41, J42, J43, J44, J45, J46, J47, J48, J49, J51and J55weresignificantly higher than that of the CK (Beilian5) while sowed in spring in Lintao county, sotheir grain can be used as the forage. The grey correlation analysis showed that, whileconsidering the hay yield and nutrition, including crude protein, acid detergent fiber and neturaldetergent fiber, of these lines, better performance observed in the lines J2, J44, J21, J28, J10andso on, therefore, further examination should be paid on these lines.For the59triticale lines, their Hay yield and seed yield were different while sowed in springand autumn. In order to obtain high Hay yield, the line J3, J6, J9, J15, J20, J31, J34, J37, J50, J53,J56, J58and CK could be sown in spring or autumn. J4, J5, J7, J13, J16, J17, J18, J19, J23, J32,J38, J49, J54and J57should be sowed in autumn; the others had better performance sown inspring. While for the seed production of these material, lines J5, J8, J22, J23, J37, J43, J44andJ45should be sown in spring and the others had good performance for the autumn sown. According to the hay productivity and quality of53triticale lines sown in Maqu, lines J57, J32,J33, J29, J28, J41, J44, J54, J59, J26, J31, J27, J58, J24, J45, J43and J21had better performancethan that of the CK and they can be used as forage cultivated in this region.The results for the suitable planting area of the53triticale lines showed that, lines J14and J16could be sown both in Maqu and Lintao, lines J2and J10were suitable to be sown in Lintao, andthe others had better performance while sown in Maqu. |
