| Prunus mume owns outstanding ornamental features in China, and it is one of the longest lived specie of the flowering trees. It is world-famous as a kind of landscape plant with early bloom and colourful flowers, and aslo used as a kind of medicinal and edible material frequently. Recently, because of the unique ornamental and economic value, Prunus mume is popular among urban greening.In this paper, using stems with buds of Prunus mume as mainly test material, the influence of different sampling time and basical medium and different hormone combinations on explantation in vitro propagation was studied. Three cultivars,’Xiao Lu’e’(Lu’e Group),’Gong Chun’(Gong Fen Group),’Fuban Hangxiang’(Huang Xiang Group), were picked from six ones, which were used to conduct preliminary culture in vitro. The main results were as the follows:1. The results showed that the most suitable sampling time for explantation was between late April to early June, and the budding rate was more than80%, at the same time, the polluting rate had been reduced significantly compared to other sampling time, for example, from July to November.2. The best initial medium exhibited different among Prunus mume cultivars. The best one of ’Xiao Lu’e’cultivar,’Gongchun’,’Fuban Huangxiang’ was1/2MS+1.0mg/L6-BA+0.5mg/L KT+0.1mg/L NAA,1/2MS+0.5mg/L6-BA+0.5mg/L KT+0.1mg/L NAA,1/2MS+1.0mg/L6-BA+O.1mg/L NAA, respectively, and the budding rate of them were87%,94.4%,70%, respectively.3. The factors influencing regeneration of Prunus mume were analyzed, such as basic medium, carbon source, cytokinin and auxin. The results showed that the optimum basic medium of ’Xiao Lu’e’,’Gong chun’ was Q&L, while WPM was that of’Fuban Huangxiang’; Three percent sucrose was benifit to explants growth and shoot multiplication, compared to glucose, maltose and sorbitol. The problem of chlorisis and STN (shoot-tip necrosis) could be solved by adding0.5g/L AC, which can guarantee the quantity and the quality of the adventitious buds during the multiplication cycle of about50days; the best shoot proliferation medium of three cultivars was Q&L+0.5mg/L6-BA +0.05mg/L NAA, Q&L+0.5mg/L BA+0.5mg/L KT+0.05mg/L NAA, WPM+0.5mg/L6-BA+0.1mg/L NAA, respectively, meanwhile, the multiplication coefficients reached4.27,2.54and2.24, respectively.4. Generally, Prunus mume reproduced by grafting and cutting. However, it was difficult to root in vitro. So the experiment that supplied different concentration of IBA and NAA for rooting medium, showed that both IBA and NAA were negative to root on three cultivars. The reason was that NAA and IBA induced large callus, so the plantlets produced rare roots, and rooting rate was only36%. When the plantlets was dipped into200mg/L-500mg/L IBA solutes in few seconds and cultured in1/2Q&L, the rooting rate was more than80%, and the average root length was3.81cm, meantime, the survival rate was higher. |
