Endotoxin is a major compoment and the pathogenic ingredient of the cell wall ofGram-negative baC teria. Under physiological conditions due to less content of endotoxin inanimal body not and the body’s own defenses will not cause a harm to the animal organism. Whenthe external environment changed suddenly the endotoxin levels rise a lot and the body’s immunefunC tion disorder can cause injury to the animal body.With the development of global warming intensive chicken farming pattern, more and moreattention to the harm of poultry industry on which heat stress is caused by high temperature insummer. Broilers for high temperature especially sensitive to the outside temperature when theexternal environment over the broiler comfort zone limit will cause heat stress on broilers.Endotoxin plays a importent role in the mechanism of injuries caused by heat stress in broilers,therefore in the process of treatment of heat stress should first consider removing or reducing theharmful effeC ts of endotoxin on animals.The test by copying the broilers acute heat stress model in order to investigate the endotoxineffeC ts renal injury of broiler and the mitigation effeC ts of pre-VC addition onendotoxin-induced renal injury after acute heat stress.This test through clinical observation、blood cell count、limulus test、physiology andbiochemistry、histopathological and molecular biology deteC tion methods are observed and testedsuch as the clinical change、the number of blood cell、serum endotoxin level、renal index andserum eleC trolyte concentration、organization slice of intestinal and renal and endotoxin receptorTLR4mRNA expression in renal indicators of changes for observation and deteC tion. In order toinvestigate the endotoxin that lead to injury and pathogenic mechanism on renal in broilers causedby acute heat stress.Routine blood test result: The number of red blood cells are slightly increased in the earlyexperimental of heat stress in broiler, along with the extension of heat stress time the number ofred blood cells began to decline and the middle late significantly reduced. The number of whitebloods are significantly reduced in the early of heat stress and along with the extension of heatstress time the number of white blood cells are significantly elevated. Between the heat stressgroup and VC broiler Red and white cell count are no significant during the heat stress reaC tion.Limulus test result: The serum of endotoxin content is significantly increased in the trial group of heat stress in broilers, along with the extension of heat stress time endotoxin content continue torise, the endotoxin is almost seven times Compared with the control group in the trial group ofheat stress in broilers. Between the heat stress group and VC broiler endotoxin content are nosignificant during the heat stress reaC tion.Physiological and biochemical test result: The serum of creatinine content increasedsignificantly in the trial group of heat stress in broilers, along with the extension of heat stress timethe increase is more obvious; Urea nitrogen is not significantly increased in the trial group of heatstress in broilers, with the extension of heat stress time it increases significantly; The eleC trolytesinculding k+、Na+Concentration have different change during the heat stress in in broilers.Between the heat stress group and VC broiler there are no significant during the heat stress reaCtion.Pathological test result: The test group are varying degrees of intestinal tissue congestion、intestinal villi disorganized、epithelial cell shedding、reducing the number of intestinal glands、central laC teal disappear inflammatory cell infiltration phenomenon at the differert heat stressreaC tion; The renal are occur interstitial congestion、 expansion、 tubular epithelial cellproliferation、wall swelling、luminal narrowing、Glomerular shrinkage cracking of inflammatorycell invasion and so on during the different heat stress reaC tion.Molecular biology test result:The test group of endotoxin receptor TLR4mRNA expression inrenal tissue is rising in the trial group of heat stress in broilers, The expression of test group TLR4is Continue to rise with the extension of heat stress time. TLR4mRNA expression in renalvolume was positively associated with heat stress time.The number of red cells are first increased and then decreased while white cells firstdecreased and then increased in broiler under heat stress condition. Its causes destroyed immunesystem and reduce the ability of resistance to endotoxin. The ability of intestinal barrier toendotoxin is reduced due to the disorders of intestinal barrier damage and immune system disorder,resulting in a large number of endotoxin shift into the bloodstream through the damaged intestinalbarrier formation of endotoxin blood disease and systemic inflammatory response syndrome. Withthe prolonged heat stress the damage degree of endotoxin in animal body intestinal endotoxemiaresulted in upregulation of renal endotoxin receptor TLR4expression of mRNA lead to renaldamage.
