The Study On The Laws Of Decreasing And Increasing Of Mast Cells In The Uterine Wall Of Rats At Oestrous Cycle

Rat a common experimental animal in medical, which often is widely used invarious medical experiments, especially in the breeding research for its short breedingperiod.Mast cell is a kind of important local immune effector cell of rat uterine wall,and plays an important role in regulating local immune level. By observing thevaginal tissue smear in rats of sexual maturation period, we determined thecharacteristics of rats vaginal tissue smear among the proestrus、estrus、metestrus anddiestrus to provide a reliable and simple method to judge for clarifying the rat’s stageof estrus;We used toluidine blue staining method to dye the uterus of rats in differentestrous cycle, studied the change regular of distribution number and form of MC inuterus of rats from the Angle of the morphological.,by using fluorescencespectrophotometer measured Histamine (Histamine) content the secretion of the mastcells in different estrous cycle of female rat uterine wall,to provide morphologymaterials and theoretical basis to further study on the relations between the MC andanimal uterus local immune.Experiment one study of distinguishing the different stages of rats in estrus cycleBy doing the vaginal tissue smear,microscopy then observe the changes of thecells of smear of20only6months of age of female rats,we determined thecharacteristics of vaginal tissue smear of rats in different stages of the estrouscycle.The results showed that:in diestrus,white blood cells give priority.Inproestrus,there are mainly nucleated epithelial cells.In estrus,there are mainlykeratinized epithelial cells.In metestrus,the number of keratinized epithelial are morethan that of epithelial cells and white blood cells.Experiment two study of the distribution of mast cells in the inner wall of the uterusin rats at different stages of the estrus cycleBy vaginal tissue smear method to distinguish the estrous cycle in rats atdifferent stages, the estrus cycle different stage of rat were executed cervical dislocation to death, application routine paraffin section and toluidine blue dyesolution dyed, we observed the dynamic change regularity of the distribution,morphology and quantity of the MC in rat uterine wall estrus cycle at differentstage.The results showed that in metestrus, rat uterine horn at the grass-roots level hadthe largest number of mast cells;The change rule of each part of the uterus in eachperiod were as follows: Metestrus (ME)>Proestrus (PE)> Diestrus(DE)> Estrus (E).Experiment three the changes of HA in the uterus of rat in estrous cycleBy the use of fluorescence spectrophotometer，we observed the change rule ofthe medium HA from the MC of the wall of the uterus of estrus cycle differentstage.The result shows: the change of HA from MC was asfollows:Metestrus>Proestrus>Diestrus>Estrus,the content of HA in uterine horn hadsignificant difference between each groups (P<0.05), but that of the cervical of uterushad no significant difference (P>0.05) in the body.
