Studies On The Anti-tobacco Mosaic Virus Activity Components Of Streptomyces ZX01Broth

Plant virus disease is very general that difficult to prevent and cure onagricultural production, therefore has brought great losses to crop yield.Microorganism is an important resource treasure for us to develop inhibitors of plantviruses, it is an important way to develop inhibitors of plant viruses frommicroorganisms. Streptomyces ZX01was isolated from the wetlant of kanas lakeXinjiang Province by Northwest A&F University Research&Devlopment center ofBiorational Pesticide. By the Previous studies of our lab, Streptomyces ZX01fermentation broth has significant inhibitory activity to TMV. To determine theanti-TMV ingredient of Streptomyces ZX01fermentation broth, we have usedTMV-tobacco as material, based on the activity tracking, combining Macroporousresin, silica gel, C18phase silica gel, Sephadex LH-20Column chromatography toseparate the anti-TMV virus activity components. The main results are obtained asfollows:1. Used macroporous adsorption resin column chromatography to remove thesalts and small molecules that was not easily adsorbed in the fermentation broth ofStreptomyces ZX01. Measured the inactivation of each elution fraction on TMV invitro. The result shows15%methanol section,30%methanol section and50%methanol section show very highly passivation activity to TMV in vitro at theconcentration of5mg/ml. The inhibition rates were96.09%,99.91%and96.27%.2.Four compounds were isolated in ethyl acetate extract from50%methanolmacroporous resin elution section of broth of streptomyces ZX01. Those chemicalsinclude:2-Furancarbosylic acid, Sodium acetate trihydrate,1,3-butanediol,2,3-butanediol.3.Using Macroporous resin, silica gel,C18phase silica gel, Sephadex LH-20Column chromatography, based on the activity tracking, isolated four compounds anda relatively pure section named C from30%methanol elution fraction after macroporous resin. The four compounds are Alanine, Isoleucine, Leucine, MethylL-pyroglutamate. Thin-layer chromatography methods, cascading mass spectrometry,nuclear magnetic resonance were used to quantitative analysis for C. Initiallyidentified the main activity compound in the fermentation broth ZX01is polypeptide.The molecular weight of main compound in section C is814,contains(Leu/Ile-Leu/Ile-Leu/Ile-Leu/Ile)—.The anti-TMV compounds obtained from broth of Streptomyces ZX01arecomplex,and big polar. Determine the structure of the active compound, pending thefurther study.
