In this study,4-20week-old of Jinlan Cashmere Goats were selected to be the experimental animal. The superovulation, cell culture technique and real time-PCR were performed to investigate the optimum superovulation method, the cultivation system of oocytes in vitro maturation culture of Jinlan Cashmere Goats kid and to determine the different expression profile of maturation genes and their receptors mRNA in immature oocytes and mature oocyte. Our study was designed to research how to produce more good quality embryos to better protect its excellent germplasm resources of Jinlan Cashmere Goats, and to provide a theoretical basis for further study of its genetic breeding process. The main results are as follows:(1)The optimum superovulation method of Jinlan Cashmere Goats kid was obtained:6-8weeks old was the suitable superovulation weeks. Difference between domestic and imported hormone was not significant (P>0.05).The optimal solution of superovulation was FSH+PMSG, injecting2ml FSH for two consecutive days at8:00am and5:00pm and400IU PMSG when the last injection of FSH. There was no appreciable impact to their adult reproductive performance after ovum pick-up of living kid.(2) The cultivation system of oocytes in vitro maturation culture was established:The oocyte maturation rate can be significantly improved by adding100μg/ml glutamine to the culture medium (P<0.05).After addition100μg/ml glutamine, adding50μg/ml or100μg/ml IGF-I markedly increased the rate of occytes maturation (P<0.05).(3) Maturation genes and their receptors mRNA appeared to be differentially expressed after oocytes maturation:BMP6,BMPRⅡ,TGF βRI and BMP RIB were significantly up-regulated (P<0.01); Ptgs2was remarkably down-regulated (P<0.01);There was no significant difference to BMP15and GDF9(P>0.05).
