This thesis contains two relatively independent parts, i.e. synthesis of chiral amines by bioconversion from oximes and metabolism of acetophenone oxime by microbes from soil.About400strains were isolated from soil samples collected in several areas, employed acetophenone oxime as the screened pressure. No strain could catalyzed acetophenone oxime to a-phenylethylamine detected by HPLC analysis.94strains were isolated from soil.8strains which could metabolize acetophenone oxime were screened by means of HPLC, among which the highest activity was obtained by strain F-44. The metabolite was identified by GC-MS which indicated that the metabolite is acetophenone.HPLC analysis indicated that the sub metabolite of acetophenone oxime was a-phenylethyanol and the metabolite of benzaldoxime was benzoic acid by strain F-44.The effects of O2, NAD(P)H, metal ions and inhibitor on the metabolism of strain F-44were studied. The results showed that the enzyme activity is60.73U/mg under anaerobic conditions. The metabolism was significantly inhibited by O2, Cu2+, Fe3+and EDTA while accelerated by NAD(P)H, Ba2+, and Mg2+.
