The utilization of defatted silkworm pupae and Microbial ecological agent in diet ofjian carp(Cyprinus carpio var. jian) was invested.This experiment was conducted to study theeffects of fish meal(FM) replacement by defatted silkworm pupae(DSP) and dietary EM on thebacterial community structure,growth performance, body composition of Jian carp(Cyprinuscarpio Var.Jian). The experiment results showed as follow:(1) This experiment was conducted to study the effects of fish meal(FM) replacement bydefatted silkworm pupae(DSP) on the bacterial community structure in the intestinal of Jiancarp(Cyprinus carpio Var.Jian) using a culture-independent method of16S rDNA PCR-DGGEfingerprinting.Five isonitrogenous(36.8%crude protein) and isoenergetic(5.5%crude lipid) dietswere formulated as follows:one basal diet with10%FM (DSP0),three diets with25%,50%,and75%FM replacement diet (DSP25,DSP50,DSP75),and two diet with0.7%crystalline lysinesupplementation in the50%,75%replacement diet(DSP50-Lys,DSP75-Lys). A total of324juvenile Jian carp with an average body weight of(15.30±2.97)g were randomly assigned into6groups with3replicates per group and18fish per group.The results showed as follows: By the analysis of PCR-DGGE fingerprint,we found thatgroup of DSP0、DSP25、DSP50、DSP50-Lys、DSP75、DSP75-Lys had19,15,11,12,8and12bands, respectively, which can be identified,and the dominant bacteria were1~2bands. thesimilarity coefficient of DGGE fingerprint of DSP0bacteria and the other group were50.7%,50.6%,69.0%,42.4%,66.7%. The major bands of PCR-DGGE fingerprinting were recoverd,cloned and sequenced,19sequences were obtained. By NCBI database homology analysis resultsshowed that bacterial communities from grass carp contents mainly wereBacilli,Spirochaetes,Betaproteobacteria,Clostridia,Actinbacteria,Gammaproteobacterial,Cyanobacteria,Bacteroidetes,Fusobacterial, including15sequences which were uncultured bacterial.Inconclusion, replacement of graded dietary fish meal protein with defatted silkworm pupae mealmade a significant impact on the bacterial community structure in the intestinal of Jian carp,andusing Lysine can reduce this impact,this may help to lay the foundation for the further applicationof compound feed of carp.(2)A feeding trial was performance to investigate the effects of dietary Microbial ecological agent(Bacillus subtilis and Lactobacillus sp.) on growth performance,body composition,intestinal micmflora,in cyprinus carpio var. jian with initial body weight of45.12±1.97g.132individuals of Jian carp were randomly divided into two treatments with three replicate tanks at arate of22fish per tank,and fed the basal diet in the control group,and the basal diet supplement with EM in the treated groups for60days.The final body weight (FBW), weight gaining rate(WGR), specific growth rate (SGR), feed intake, protein efficiency ratio (PER) of fish in thetreated group were significantly higher than those in control groups(P<0.05),as well as the CFand IFI(P<0.05);and feed conversion ratio (FCR) was significantly lower than those in controlgroup(P<0.05). The IFI in the treated group were significantly higher than those in controlgroups(P<0.05).No significant difference of CF、 RGL、HIS and SI were found in thesegroups(P<0.05),as well the content of crude protein、moisture、crude lipid、crude ash (P>0.05).The content of C20:1n-9、C18:3n-3and∑MUFA in muscle of treated group were significantlylower than those in control group(P<0.05),and the contents of C22:4n-6、C22:6n-3(DHA) inmuscle of treated group were significantly higher than those in control group(P<0.05). Therewas signficiantly higher number of Bacillus sp.(P<0.05)and Lactobacillus sp.in the treatedgroups(P<0.01),while there was significantly lower of E. coli than that in the control group(P<0.05),indicating that supplement of EM in diets led to increase in the improvement of the growthperformance and intestinal microflora in Jian carp cyprinus carpio var.
