| In order to research the mechanism of nitrogen on tobacco qualityformation and metabolism characteristics in Dali and Qujing, Honghuadajinyuan andK326were planted in same nitrogen fertilization mode in two areas. Key enzymes andgene of metabolism of Flue-cured Tobacco were tested to analysis the difference ofmetabolism between Dali and Qujing; Chemical components were tested to analysisthe difference of style of flue-cured tobacco between two areas. The main results areas follows:1. Difference of carbon, nitrogen and secondary metabolism of K326andHonghuadajinyuan between Dali and QujingResearch of carbon and nitrogen metabolism of K326between Dali and Qujing,showed that: nitrogen metabolism exuberant period started at50d in Qujing, Dalistarted at60d, more carbon skeleton turned into carbon metabolism in transformationprocess between carbon and nitrogen metabolism in Qujing; and Dali tobaccocarbohydrate accumulation period, mainly concentrated in the early period, Qujingtobacco leaf carbohydrate accumulation and distribution in the whole growth periodof tobacco, Qujing tobacco needs a lot of irreversible carbohydrate in the mediumterm, these factors caused the Qujing tobacco entered the era of technology maturelater than Dali tobacco.Research of secondary metabolism of K326between Dali and Qujing, showedthat: Because of the difference of carbon and nitrogen metabolism of flue-curedtobacco in Dali, Qujing than tobacco entered earlier phenylpropanoid metabolismexuberant period, polyphenol accumulation were correspondingly higher than thelatter. Research showed that the differences of accB and DXR gene expression, Dalitobacco terpenoid metabolism and fatty acid metabolism are mainly concentrated inthe early period, Qujing tobacco is distributed in the whole growth period.Research of chemical components of K326between Dali and Qujing, showedthat: Dali tobacco leaf of lutein and beta carotene content higher than that of Qujingtobacco, and content of total sugar, sugar and starch had little difference, content oftotal nitrogen and nicotine of flue-cured tobacco in Qujing is higher than Dali tobacco.Research of metabolism and quality of Honghuadajinyuan between Dali andQujing, showed that: The difference of metabolism and quality of Honghuadajinyuanbetween two areas is same as K326, and key enzymes and gene of metabolism ofK326and Honghuadajinyuan in same place changed in similar trends. These resultsshowed that: Ecological conditions play more important role on style of flue-curedtobacco than varieties and nitrogen fertilizer.2. Difference of carbon, nitrogen and secondary metabolism between K326andHonghuadajinyuan in same areaResearch of Metabolism and Quality of tobacco between K326andHonghuadajinyuan showed that: nitrogen metabolism of Honghuadajinyuan is moreexuberant than K326in the late period, the carbon flux distribution has a certaindifference between two varieties. In the early stage, carbon source of K326flowedmore to the reversible carbohydrate direction, while in the later stage carbon source ofHonghuadajinyuan flowed more to the irreversible carbohydrate direction; K326hadlarger accumulation of sucrose and stronger transport activity of sucrose in early stage,and sucrose accumulation period of Honghuadajinyuan big tobacco mainlyconcentrated in the late. The experiment results also show that the content of rutin inK326tobacco leaves was higher than that of Honghuadajinyuan, and camphorphenanthrene alcohol based-3-rutinoside content is lower than the latter; polyphenoloxidation in K326is higher than Honghuadajinyuan in early stage, benzylisoquinolinealkaloids synthesis metabolism is also corresponding higher. Chemical compositionsof two varieties also had the certain difference, the nicotine content in K326is lowerthan Honghuadajinyuan, and sugar nicotine ratio is higher than the Honghuadajinyuanleaves.3. Difference of carbon, nitrogen and secondary metabolism between differenttreatments of nitrogen in same areaResearch of nitrogen on metabolism and accumulation of tobacco in same place,showed that: Application of nitrogen can improve tobacco NR and GS activity in theearly growth period, make nitrogen metabolism more exuberant in early growthperiod, and more nitrogen metabolite accumulated, provide carbon skeletons moreinto the carbon metabolism in transformation process between carbon and nitrogenmetabolism; Application of nitrogen can increase leaf PAL activity, enhanced phenylpropanoid metabolism, promote polyphenol synthesis; nitrogen can prolongleaf fatty acid biosynthesis metabolism exuberant period and synthetise more fattyacid. At the same time nitrogen also has a certain effect on nitrogen accumulation intobacco. Nitrogen application can increase flue-cured tobacco in lutein and betacarotene content, promote the synthesis of carotenoids; at the same time nitrogen canreduce the content of total sugar, sugar and starch in tobacco leaf, and increasedpotassium content in tobacco leaf, make chemical composition of tobacco leaf morecoordination. These result shows that flue-cured tobacco applied with nitrogen hasmore exuberant carbon, nitrogen and secondary metabolism in its growing up thanwhich applied without nitrogen. |
