By PCR and RACE technology, we cloned the Sucrose Transporter Gene PsSUT1from theleaf of Paeonia suffruticosa cultibvar ‘Huhong’, expression pattern of the PsSUT1in leaf andpetals of tree peony at different developmental stages was analyzed by RT-qPCR. We analyzedthe gene expression in the process of peony flower, which provided a base for the research onmolecular mechanisms and transport of carbohydrate during flower development of peony. Theresults of this research are as follows:1.According to the amino acids conservative district of SUT genes, we designed degenerate primerP1and P2. We cloned the SUT genes(PsSUT1), the full length cDNA of PsSUT1is1864bp, openreading frame is1557bp, encoding a protein of519amino acids, it belongs to the GPH proteinfamily, the GeneBank registration number is KC542395.2. The characteristics of PsSUT1meet SUT1subfamily proteins, it has12typical transmembranestructure, and it is membrane combined protein. According to homology analysis, PsSUT1shareshigh homology with Medicago truncatula(AFM28284), Hevea brasiliensis(CAM34330), Populustrichocarpa(ADW94613), Vitis vinifera (AAF08331). The analysis of the genetic evolution showthat it has close genetic relationship with walnut sugar transporters of SUT1subfamily proteins, soPsSUT1was classified as SUT1subfamily proteins.3.The analysis of the expression of peony PsSUT1gene in four growth periods shows that,PsSUT1expressed in roots, stems, leaves and petals of tree peony, especially in sugaraccumulation parts such as stems and roots the expression quantity is higher, but it is lower inpetals. The expression level in stems was10.9times than that in petals at flowering stage.Expression level of PsSUT1at flowering stage was the highest, which was379%higher than thatat peach periods.From what has been discussed above, the PsSUT1which cloned from the peony is themember of peony sucrose transporters gene families. They are closely related with peony sugartransport and accumulation process, which provided a base for the deeply research on molecularmechanisms of metabolism and transport of carbohydrate during flower development of peony.
