Objective:In order to diminish Capsaicin adverse drug reaction, we prepared Capsaicin delivery system. The delivery system of drug can release drug be more durable, reduce the frequency of administration, reduce peak and valley phenomenon of PLLAsma concentration, provide a smooth and lasting effective blood concentration, reduce toxicity, and improve safety and effectiveness of drug.Methods:In this paper, poly-L-lactic acid (PLLA) was used as the carrier and for capsaicin was the model drug. Polylactide (PLLA)/Capsaicin composite membranes with nanofiber structure (mats Ⅰ),"bead on string" structure (mats II) were prepared by electrospinning. A dense membrane (mats Ⅲ) was also prepared by solution casting method. Nanostructures was observed by SEM. The release of drug was detected by UV spectrophotometry. The effects of microstructure and drug loading (5wt%,10wt%and15wt%) on the releasing rate were studied.The following results were obtained:1. The PLLA nanofiber membrane were successfully prepared by electrospinning method. The properties of PLLA nanofiber were characterized by SEM、DSC、shape memory properties, the results show that10wt%PLLA nanofibers’diameter are uniform, and the nanofiber are smooth. So,1Owt%PLLA is the best test condition.2. The PLLA/Cap nanocomposite composite membranes and bulk composite membranes were successfully prepared by electrospinning method and solution casting method.3. PLLA/Cap composite nanofiber membrane had some slow-release effect for capsaicin by in vitro release test. Both the microstructure and drug loading of the PLLA/Cap membrane played important roles in the releasing of capsaicin. When the drug loadings were5wt%or10wt%, the order of the capacicin release rate is:Mats I> Mats II> Mats III. With the increase of capsaicin loading, the durg release rate increases, and the differences between the composite membranes decrease. When the composite membranes’drug loading reach15wt%, the effect of drug loading on the drug release rate become more remarkable, and the drug release rates of the three composite membranes become more close to each other.4. Drug release test of capsaicin form PLLA/Cap composite film indicate that the higher drug load, the more cumulative release of capsaicin. The cumulative release amount of drug-loaded5wt%、10wt%、15wt%are60%、90%'100%in120h, respectively.Conclusion:The release rate of capsaicin in the composite membranes at the was affected by the microstructure of membranes and drug loading at same time.
