Polygonum viscosum is a wild species with ornamental, medicinal,edicinal value, and which has special aroma. The wild Polygonum viscosum from Mao’er Mountain in Shangzhi of Heilongjiang province were the test material of this thesis.Explore fast and effective propagative approach in two ways:tissue culture and seed germination and preliminary study the Polygonum viscosum oil extraction technology with the method of steam distillation. The mainly research results are as follows:1. The characteristics of Polygonum viscosum seed:The seed length was2.50-3.16mm, and the diameter was1.82-2.16mm. The1000-grainweight was4.81g; the average viability was85.56±1.11%. The weight of seed coat, endosperm and embryo respectively account for40.29%,55.61%and4.01%of the whole seed weight. By observing the structure of the embryo, the morphology of the embryo is fully developed.2. One reason of Polygonum viscosum seed difficult to germinate was testa has mechanical impediment, so the embryo can not break through the seed coat growing normally. The seed sterility test suggested that the radicle and cotyledons did not exist dormancy, but its germination needs nutrition.3. When the Polygonum viscosum seeds at25℃,1900-20001x light conditions, there are the highest germination rate and germination potential, so it is the best conditions for seeds germination. Only when the mechanical damage and exogenous hormone treatment simultaneously affect to seeds, it can be improve the germination rate. Concentrated sulfuric acid treatment improved the seed coat permeability, but the effect is somewhat less than the mechanical damage. Three kinds of exogenous hormones GA3、6-BA and NAA treatment the mechanical damage seeds, all improved the germination rate, which GA3best. After the concentrated sulfuric acid treatment and then200mg·L-1GA3soaking, then laminated30d, that can improve the germination rate, and seedling growth robust. After32kinds of treatment methods, the mechanical damage seeds through soaking in200mg·L-1GA348h, that germination rate and germination energy were the highest, respectively,58.00%and31.33%, average speed germination was4.36%, seeds germination results best.4. The stems with axillary buds, leaves,petioles and stems of Polygonum viscosum were as explants, establishing the tissue culture and rapid propagation system of Polygonum viscosum after the first generation of induction, subculture and rooting transplanting. The test studied the influence of different plant growth regulators to the tissue culture of Polygonum viscosum. The results of this study showed that treated with5%NaClO solution for10min was the best way to disinfection of the stems with axillary buds and treated with0.1%HgCl2for4min was the optimum explants disinfection of leaves of Polygonum viscosum. The optimum axillary bud induction culture medium was MS medium added with2.0mg·L-16-BA purine and0.1mg·L-’NAA, having the highest induction rate and shoots stout; the optimum proliferative medium for axillary buds was MS medium added with2.0mg·L-16-BA,0.3mg·L-1NAA acid and O.5mg·L-1KT, having the highest multiplication times simultaneously differentiation buds thick and green; the optimum seedling effect medium for axillary buds was MS medium added with1.0mg·L-16-BA,0.1mg·L-1NAA acid, which can be directly used for rooting; the optimum root induction culture medium was1/2MS medium supplemented with0.7mg·L-1NAA. Induced roots were longer, more fibrous roots. Plants grew well after being transplanted.5.The best extraction crafts of Polygonum viscosum’s essential oils by Steam Distillation was:with extraction of3.5h,350ml water,2%of NaCl. The oil content of the leaves was higher.Volatile oil by GC-MS analysis of the fragrance exudes fruity flavor mainly is2-methylbutyl-2-methyl butyrate, which easter-based oil in the region of low boiling compounds contained various esters are unique.
