This experiment was conducted to investigate the Effects of Nephrotropic Strains ofInfectious Bronchitis Virus on Chicken Clinical pathology and Expression of Relatedgenes.240hy-line variety brown were randomly divided into virus group and control groupin35days, three repetitions were set in each group, each repetition involved40chickens.In day0, virus group was arranged to accepted the artificial nasal drip according to theresults in ELD50of embryos(10-5/0.2ml),0.2mL of each chichen, however, the controlgroup remained blank. In respective in8th15thand22thdays, six birds randomly selectedfrom each group were sampling, blood sampling and separation serum were collected andmeasured the function of liver and kidney、electrolyte indicators、antioxidase enzyme index.At the same time, collected liver and kidney in order to determining the expression ofXOD and apoptosis related genes Bcl-2、Bak1、P53mRNA. The results are as follows:(1) Compared with control group, in8thday, Ca2+、P+and K+increased significantly(P<0.05); in15thday, yet P+increased significantly(P<0.05); in22th day, Ca2+decreasedsignificantly (P<0.01), P+and K+declined considerably(P<0.05orP<0.01).(2) Compared with control group, in8th15thand22thday, UA increased significantly(P<0.05); in15thand22thday, CR increased significantly (P<0.05).(3) Compared with control group, in8thday, ALT、AST and5-NT increasedsignificantly (P<0.05); in15thday, TP、ALB、GLB、ALT、AST and GGT significantincreased (P<0.05).In22thday, GLB and LDH showed an obvious decreasingtendency(P<0.05).(4) Compared with control group,in8thand15thday,XOD and MDA increasedsignificantly (P<0.01), but GSH-PX T-AOC and T-SOD showed dramatic fall (P<0.05orP<0.01); in22th day, XOD showed a increased tendency (P<0.05).T-AOC and GSH-PXdecreased(P<0.05).(5) Compared with control group, in8th15thand22thdays, XOD mRNA geneticexpression in kidney and in liver had a much higher level than expression of control group(P<0.05orP<0.01).(6) Compared with control group, in8th15thand22thdays, BAK-1and P53mRNAexpression in kidney increased significantly (P<0.05or P<0.01); in8thday BAK-1andP53mRNA expression in liver increased significantly (P<0.05).in8thday BCL-2mRNAexpression in liver and kidney raised dramatically(P<0.05).In conclusion: Nephrotrophic strains of IBV could lead to electrolyte imbalance,antioxidant function disturbance and kidney dysfunction and hepatic dysfunction.
