Co-relationship Analysis Of Apoa-Ⅰ Gene、apoB Gene Polymorphism With Fat Metabolism In Jiangxi Indigenous Chicken Breeds

Abstract/Summary:
In this experiment,50Chongren spotty chicken, Yugan chicken, HY-line layer, Greenshell layer, Anyi Wahui chicken,Ning-du yellow chicken and Taihe silky fowl were used astest materials and primers were designed according to the sequence of apoA-Ⅰ and apoBgene. The single nucleotide polymorphisms of apoA-Ⅰ and apoB gene were detected byPCR-DNA direct sequencing and its genotypes were analyzed by read-through of DNAsequence along with the identification of its peak chart, which aims to explore therelationship between apoA-Ⅰ、apoB gene polymorphism and fat metabolism in indigenousbreeds of chickens. Results are as follows:1.In apoA-Ⅰ, one SNP was found and a single base was transformed (A→T) on163bp spot of the initation codon ATG, leading to three genotypes: AA,AB and BB. ABgenotype was the superior genotype in Yugan chicken and BB genotype was the dominantgenotype in Green shell layer, Anyi Wahui chicken,Ning-du yellow chicken; and allelegene A was the superior allele in Yugan chicken and Taihe silky fowl, allele gene B was thesuperior allele in Green shell layer, Anyi Wahui chicken,Ning-du yellow chicken. Byanalyzing apoA-Ⅰ gene polymorphism, all kinds of tested chickens’s polymorphic locuswere moderate, and they were in Hardy-Weinberg equilibrium except Green shell layer.2.In apoB, one SNP was found and a single base was transformed (T→G) on123bpspot of exon26, leading to three genotypes: TT, TG and GG..TT genotype was the superiorgenotype in Chongren spotty chicken and Taihe silky fow, and TG genotype was thesuperior genotype in Yugan chicken, Green shell layer, Anyi Wahui chicken,Ning-duyellow chicken.; and allele gene T was the superior allele in all kinds of tested chicken; Byanalyzing apoB gene polymorphism, all breeds of tested chickens’s polymorphic locuswere moderate, and only HY-line layer, Yugan chicken and Anyi Wahui chicken were inHardy-Weinberg equilibrium.3.The liver fat rat, the abdominal fat weight and the abdominal fat rat of Chongrenspotty chicken and Yugan chicken which contain allele gene B were significant higher(P<0.05)than HY-line layer, it carried out that allele gene B was significantly correlatedwith the abdominal fat weight and the liver fat rat in in Yugan chicken and Chongrenspotty chicken. Both of the abdominal fat weight and the abdominal fat rat of Yuganchicken was significantly lower(P<0.05) than Chongren spooty chicken which containsallele gene G, it showed that allele gene G was significantly correlated with the abdominalfat weight and the abdominal fat rat in Yugan chicken and Chongren spooty chicken.All the results obtained in the present study confirmed that apoA-Ⅰand apoB genewere a significant association with fat metabolism in Jiangxi indigenous chicken breeds;and apoA-Ⅰand apoB gene were the potential marker for use in molecular marker-assistedselection program related with fat traits in chicken.
