Cotton fiber as one of the most important natural fiber raw materials in the world,its importance is self-evident in the textile industry, improving the quality of thecotton fiber become a primary objective of the cotton. With the continuousimprovement of gene cloning technology, a large number of advantage or specificexpression genes in cotton fiber development period have been cloned. In thisexperiment we chosen seven reported cotton fiber development advantage expressedgenes (GhACT1, GhCAD1, GhCCR1, GhExp1, GhExp2, GhCIPK1and GhPL), firstof all, using real-time fluorescent quantitative PCR technology, analyzes the relativeexpression of specific of the seven genes above respectively in the G.hirsutum,G.barbadense and G.arboreum whose cotton fiber quality has difference in0DPA,5DPA,10DPA,15DPA,15DPA,20DPA, explicating the gene expression pattern,and investigating the relationship between the gene expression and fiber quality.Second, physical and chemical properties, the subcellular localization,n-terminalsignal peptide and protein secondary structure of these genes’ bioinformatics has beenforecast. Real-time fluorescent quantitative PCR results showed that seven geneshave high quantity expression in fiber elongation stage, but the genetic backgroundis different, seven genes expression spectrum in three varieties cotton fiberdevelopment period is also different. By comparing the relative expression ofGhExp1and GhExp2we found that the expression amount of GhExp1is much higherthan that of GhExp2, speculated that GhExp1gene is more important for fiberelongation. According to fiber differences between three cultispecies in cotton genus,analysis the relationship between gene expression quantity and the fiber quality, highexpression of GhACT1and GhExp1gene may have a certain role in the formation ofgood fiber quality, and the relationship of other gene expression level and fiberquality is not obvious. Bioinformatics prediction results show that the seven geneexpression products are water-soluble protein, secondary structure is mainly made upof spiral, folding and random coil, only GhExp1, GhExp2, GhPL1three geneexpression products has N-terminal signal peptide.
