Clenbnterol hydrochloride is a cheap drug used to reduce feeding animal fat and to increase lean role. It will have dangerous side effects for some susceptible people with obvious symptoms of poisoning, so it may have great harm to human health. Most countries including China and the European Union have banned or strictly control the use of such hormones. Original detection method for clenbuterol is time-consuming, complicated process, which need to create a more efficient, fast, and accurate method of experiment. This research is to establish the detection method by gas chromatography mass spectrometry (GC/MS) for three representative clenbnterol hydrochloride, clenbuterol, salbutamol and ractopamine.First, the extract purification effects were compared for clenbuterol, salbutamol and ractopamine at the key factors of different temperature, hydrolysis time and enzymatic fluid pH value. The method of single factor and orthogonal test of three factors was used, the ultrasound time of ammonium acetate in the pre-treatment process was adjusted and the speed of column chromatography was controlled. The results showed that the best conditions for the laboratory sample pretreatment enzymatic temperature was35℃, hydrolysis time was8h and enzymatic hydrolysis pH value was2.0. The experiments optimized the configuration methods for leacheate, the key steps for column chromatography and greatly improved the recovery rate. Second, the standard spectrum library information of missing ractopamine was estabilished, the temperature program and the analysis time were optimized, the standard curve and the detection limit were determined by mass spectrometry. Through a series of optimization measures, the experiment achieved good reproducibility and recovery. Finally, the115samples from meat products market of five cities including Nanning were not detected positive by our methods, which further confirmed the scientific and feasibility of the experiments. It also paved the way for further study.The method of the present study is the optimization process on the basis of the pre-treatment method in the original experiments. Under the premise of ensure stable recovery, the enzymatic time and the amount of the solvent were reduced, the leacheate configuration was optimized. Less chromatographic peaks of impurities after the sample procession, more reliable results and less time for detection, which meet rapid detection requirements for the most laboratories. It also offer technical support to food safety risk monitoring for the laboratories.
