Recently, two types of strain Pekin ducks have been breed by Institute of Animal Sciences of Chinese Academy of Agricultural Sciences, fat strain and lean strain. And they are significant different in growth performance. The fat strain Pekin ducks has a greater fat content than that of lean strain ducks, which is suited to be roast. Fat deposition of meat-duck main occurs in adipose and muscle tissue including skin and abdomen, and skin is the uppermost. Moderate sebum is to the benefit of cooking process, but an excess of sebum content would lower feed conversion efficiency and carcass quality. Adipocyte fatty acid-binding protein (A-FABP) is one of the important candidate genes affect fat deposition, which main expresses in adipose tissue and deposits triglyceride in adipocyte. In this study, two experiments were conducted to evaluate the characteristic of A-FABP express in different period and various tissues of both fat strain and lean strain ducks and its expression feature by eukaryoticvector construction. This study is of great significance to reveal the role of A-FABP gene in ducks’ fat deposition, and it also has a certain application value to seek one of biologicals of fatty deposits.The experiment1was conducted to evaluate A-FABP gene express characteristic in various tissues of Pekin ducks. Heart, liver, spleen, kidney, muscularstomach, duodenum, pectoral muscle, thigh muscle, sebum, abdominal fat tissues on6-week-old samples of two stain ducks were collected respectively to detect A-FABP gene expression patterns by q-PCR. The results showed:6-week-old lean strain ducks A-FABP gene express quantity in various tissues as follows:sebum>abdominal fat>muscularstomach>duodenum, heart, thigh muscle>pectoral muscle>spleen, liver>kidney, while fat strain ducks as follows:sebum, abdominal fat>muscular stomach>duodenum> heart, thigh muscle>pectoral muscle, spleen> liver>kidney; this study further demonstrated that A-FABP gene express highly in adipose tissue of6-week-old ducks, and duck A-FABP gene express much higher in sebum and abdominal fat tissues than other tissues (P<0.05);6-week-old lean strain ducks had a greater A-FABP gene expression in sebum and abdominal fat tissues than those of fat strain ducks(P<0.05); in order to investigate ducks A-FABP gene express quantity difference in different period of sebum and abdominal fat tissues, sebum and abdominal fat tissues on2,4,6,8-week-old of two types of ducks were collected respectively to detect A-FABP gene expression patterns by q-PCR. The result showed that two types of ducks A-FABP gene expression in both sebum and abdominal fat tissues increase with age increase;8-week-old fat strain ducks had the greatest A-FABP gene expression in sebum and abdominal fat tissues, while6-week-old lean strain ducks reached the maximum.The experiment2was conducted to further investigate duck A-FABP gene characteristic,pIRES2-ZsGreenl-A-FABP eukaryotic express vector was constructed, then be imbed by lipidosome Lipofecter, and to be injected to ducks’ sebum. qPCR was used to detect genes expression related to fat metabolism. The result showed the ducks which were injected A-FABP eukaryotic express vector sebum A-FABPã€PPARγã€FAS gene express on3d significant increase(P<0.05), but LPL gene express had no significant difference (P>0.05); on6d, A-FABP gene express had a increase tendency, LPL gene express significant increase(P<0.05), with no significant difference in PPARγã€FAS gene express (P>0.05). Overall, duck sebum A-FABPã€PPARγã€FAS gene express significant changed after injected A-FABP gene vector plasmid, which shows A-FABP gene express influence genes expression related to fat metabolism. |
