Rosa rugosa Thunb. is a kind of important ornament plant, and it is also a valuable natural herb and excellent landscape plant material which belongs to genus Rosa.The deciduous shrubs,which are the traditional famous flowers in our country,have a pleasant fragrance and wide variety.The stem of R. rugosa has a lot of prickles which are formed by the deformation of plant trichomes and a handful of plant epithelial cells.This leads to a lot of inconvenience in the process of field management and picking flowers.If we can cultivate a strain of R. rugosa with less prickles or even no prickles, it will greatly enhance the work efficiency of R. rugosa management and harvest. Taking the representative varieties of R. rugosa as experimental material, on the basis of prickles histological observation,we cloned a transcription factor RrTTG1which related to the prickles development, then analyzed the expression of the gene RrTTG1in different parts and different varieties of R. rugosa.The main results are as follows:1.The stem epidermis of R. rugosa’Fenzizhi’was used as plant material to study the prickles organizational form and structure by making paraffin section and the observation of scanning electron microscopy (sem).Results indicate that prickles of R. Rugosa have a level of organization.The outermost layer of protection organization has rules of closely packed cells.Close to the inside of the epidermis layer, the first and the second layers of cells which are similar to the meristematic cells of cambium morphology, are small and closely packed.Go back to the inside cells within the larger volume, arrange loosely and irregularly.There is no vascular bundles inside.All of above are similar to the structure characteristics of epidermal stem cells, which proves that prickles of R. Rugosa are a sort of multicellular trichomes without glands.2.Total RNA of’Fenzizhi’young leaves was extracted using the CTAB method, then the full cDNA sequences of RrTTGl gene which related to the prickles development were cloned by RACE method. The results for the test have shown that the full length of RrTTGl is1348bp, with the start codon, the complete open reading frame (1038bp), termination codon, the5’ non-coding region (73bp), the3’non-coding region (225bp) and poly (A) tail (12bp), which encodes346amino acids, and the Genbank No. is JX878895. By comparing the DNA sequences with publicly availably reference sequences in Genbank, RrTTG1shares90%homology with Fragaria x ananassa (JQ989287),84%homology with Rubus idaeus (HM579852) and Malns x domestica (GU173814),83%homology with Pyrus pyrifolia (HQ641374),81%homology with Prunus persica (FJ771043).The homology similarity of RrTTG1deduced amino acid sequences and Fragaria x ananassa (AFL02466), Rubus idaeus (AEI55401), Malus x domestica (AAF27919), Pyrus pyrifolia (ADU25044), Prunus persica (ACQ65867) deduced amino acid sequences are as high as97%,95%,92%,92%,92%.3.’Fenzizhi’was used as plant material to study the expression patterns of RrTTGI in different parts of R. Rugosa,Weihai wild and ’Ciguo’ were used as plant material to study the expression patterns of RrTTG1in different varieties of R. rugosa. RT-PCR and Realtime-PCR were used in all of the experiments above.lt turned out that in different varieties and different parts of R. Rugosa,the expression of RrTTGl was existed everywhere.And the highest expression was in the leaf.The second was in the stem.Pericarp and petals also have a small amount of expression.And the expression level of RrTTGl gene in prickly R. Rugosa’Ciguo’was also higher than in less prickles Weihai wild R. Rugosa.The research results show that the RrTTG1gene is possibly related to the prickles formation of R. Rugosa. |
