Seedlings from seeds are a favourite method to be used in nursery propagation of peach (P. persica L.).But it can induce characters separation easily compared with its parents. Consequently, the grwth and resistance of the seedlings were irregular. Vegetative propagation could get the uniform rootstocks, including tissue culture and cutting propagation. The asexual reproduction technology of GF677(almond X peach hybrid) has been widely used in production in Europe and America. Lots of studies about the technology have been done in China, but the’three-low’problems, low proliferation rate, low rooting rate and low survival rate of transplanting are the main limitations of seedling production, especially on the varieties in subgenus Amygdalus (L.).’Zhong Tao Kang Zhen1’peach is an excellent which resistant to replant problem breed by ZFRI, CAAS. How to propagate the rootstock fatly by vegetative propagation is the limiting factor for promotion this rootstock. In this study,’Zhong Tao Kang Zhen1’peach was used as the main material to determine the optimal culture medium which can increase the proliferation rate and the factors of cutting propagation to improve the rooting rate and survival rate of transplanting. The study is helpful to establish the system with high proliferation rate and stability for vegetative propagation technology. The results are as follows:(1) The micropropagation of’Zhong Tao Kang Zhen1’:The best period for collecting the explants is in early May, the explants collected in that time would get high germinisation rate up to90%just in3days; Axillary buds showed a higher germination rate than the apical buds. And the optimal sterilized treatments is ethanol(75%) for30s and HgCl2(0.1%) for4min. The most suitabie medium for proliferation is MS media supplemented with F mg/L6-BA, f mg/L IBA and0.2mg/L GA3and the proliferation index could up to3.53; The most suitable medium for elongation is MS media supplemented with F mg/L6-BA, j mg/L IBA and0.2mg/L GA3and the height of the shoots reach to3.9cm after cultured28d; The most suitable medium for rooting is MS media supplemented with a mg/L IBA, b mg/L IAA and the rooting propagation rate is52.5%.(The results had applied for patent)(2) The rooting on cutting of ’Zhong Tao Kang Zhen1’:The rooting rate of semi-hardwood cuttings treated by IBA2500mg/L from ’Zhong Tao Kang Zhen1’is significantly higher than other treatments, the rooting rate can reach to73.68%. The semi-hardwood cuttings have higher rooting rate in vermiculite or in mixed medium of vermiculite and sand than it in either sand or perlite. The season affected the rooting ability of rootstocks obviously. The rooting rate of ’Zhong Tao Kang Zhen1’is significantly higher that in August than in September, October and November. The rooting ability of hardwood cuttings varies in different peach rootstock cultivars.(3) The anatomical observation on adventitious roots which colleted by tissue culture and cuttings propagation of ’Zhong Tao Kang Zhen1’indicated that the incubate root primordium has not been found in culture shoots or cuttings before rooting treatment. After rooting treatment, the incubate root primordium initiates from cortex, phloem, xylem and callus cells.The incubate root primordium is induced and with multiple sites.
