Dendrobium officinale Kimura et Migo is a rare traditional Chinese medicinal herbs, have high officinal and economic value. we compare the suitable of different provenances of Dendrobium officinale planting in the central mountains of Hainan at different altitudes, and design of5different substrates to screen out the most suitable source and substrate for Dendrobium officinale cultivation in Hainan, at the same time, design the medium with different concentrations and kinds of additives,to select the most suitable medium for protocorm proliferation and rooting, cultured the immature seeds in vitro to observe the growth of seed embryo, all of these can provide scientific basis for the indusrial production. According to the variation of agronomic traits and of Dendrobium officinale at different harvest time.The main results are as follows:1.Using the same substrate cultivate the different provenances of Dendrobium officinale, we can find that "soft feet" were superior than "hard feet" in appearance and growth,100%pine bark is the most suitable culture medium for planting Dendrobium officinale in these5kinds of culture medium.2.Through the Motic B5digital microscope to observe the Dendrobium officinale germ with different in vitro cultivation days, we can find that embryo is growing very slow befor21days,embryo begin growing rapidly after21days,35days suspensor structures can be observed,42days cotyledon primordia can be observed. Dendrobium officinale protocorms have strong proliferation ability, the culture medium of1/2MS+10%coconut water+NAA0.1mg/L show the the best proliferation effect, the add of mashed potatoes will inhibit the growth of protocorm. Dendrobium officinale plantlets had the best rooting and growth on the medium basic on1/2MS macronutrients+1/2MS trace element+1/2MS ferric, high concentrations of inorganic salts will inhibit rooting and make the growth slow. Dendrobium officinale plantlets in seedling stage are not sensitive to the concentration change of NAA, IB A, IAA.3.By using of sulfuric acid phenol method to determination the polysaccharide in Dendrobium officinale’s stems and leaves. Polysaccharide content in leaves is much lower than the content in stem, Hainan growing areas with high altitude are more conducive to Dendrobium officinale stem accumulation of polysaccharides and stem dry out rate will increased with increasing altitude. Polysaccharide content in Dendrobium officinale stem are presented to three stages, grow, maintain, reduce, the highest polysaccharide content appeared in April、May、June, because of high polysaccharide content and the best agronomic traits that April is considered to be the best harvest time. Dendrobium devonianum leaf polysaccharide content is two times of Dendrobium officina’s, Dendrobium devonianum stem polysaccharide content can reach more than40%as Dendrobium officinale.Through use of HPLC to determination mannose content of Dendrobium officinale,we can find that mannose uniform distribution in stem and leaves, the influence of different altitude conditions and different planting years on mannose content was not significant, mannose content in the leaves and stems of Dendrobium officinale is slightly higher than Dendrobium devonianum. We can not detected the erianin components in Dendrobium officinale.
