Sweet potatoes are important crops of grain, feed, industry and energy.Sweet potato virus disease has seriously affected the production of sweet potatoes,causing the deterioration of its quality and degradation of variety. According to the report, at least8kinds of viruses were found in China as follows:Sweet potato feathery mottle virus(SPFMV), Sweet potato leaf curl virus(SPLCV), et al. Therefore, the establishment of rapid and efficient virus detection technology can provide theoretical basis for the understanding of different sweet potato viruses’ occurrence frequency, distribution and mixed infection as well as the prevention and control of potato virus disease.This study analysed by using serology researched the318samples of symptomatic collected from14provinces of China in2013.The results are as follows.(1)318samples of symptomatic were gathered from14provinces of China in2013, and SPFMV、SPMMV、SPC6V、SPCFV、SPMSV、CMV、SPCaLV、SPVG、SPLV、SPCSV、SPV2were detected by using NCM-ELISA and ACP-ELISA.There11kinds of viruses were detected.(2) By using NCM-ELISA and ACP-ELISA detection, the results show that SPFMV detection rate was the highest and SPV2infecting sweet potatoes occurs most commonly.(3) The two kings of virus compound infection and five kinds of virus compound infection occurred most commonly by detecting the318samples.(4) There were100kinds of mixed infection among the142samples, of which SPFMV and SPC6V are the most common ones.(5) The detections of SPFMV and SPCSV both presented positive samples with typical symptom of SPVD was widely distributed in9procinces:Sichuan, Shandong, Jiangsu, Anhui, et al.(6) Mixed infection is most easily occurred between SPFMV and other viruses.In addition,the SPCV-YU CP gene was cloned into expression vector pET-28a(+)for over-expression in prokaryotic cells by using genetic engine methods. Successfully constructed prokaryotic expression vector of pET-SPLCV CP.
