Aspergillus oryzae fermentation important industrial microbial strains, is widely used.Aspergillus oryzae has a rich cellulose degrading enzyme series, it is one of safety microbialstrains announced by the US Food And Drug Administration and American Association Of FeedCompanies, animals can be directly fed microbial products.This paper is about Aspergillus oryaze, four Aspergillus oryaze were cultured by thecellulase-producing and a new Aspergillus oryaze M2with high yield of cellulase has beenselected.The effects of different carbon source,nitrogen source and inorganic salts on thebiomass and effective metabolites of Aspergillus oryzae M2had been studied.On the basis of single factor investigation,the optimal liquid medium and culture conditionswere obtained by orthogonal experimental analysis as molasses150g/L, urea0.4%, peptone0.3%, Potassium hydrogen phosphate anhydrous0.16%, bitter salt0.07%, Sodiumchloride0.05%and Ferrosi Sulfas0.05%.In this condition, the biomass of Aspergillus oryzae was22.21g/L,which was39.60%higher than that before optimization(15.91g/L).The optimalculture conditions of50mL medium amount,culture temperature30℃,pH6.0,180r/minrotational speed and4%inoculation,In this condition, the biomass of Aspergillus oryzae was25.32g/L,which was7.43%higher than that before optimization(23.57g/L).This study was conducted to investigate the effects of Aspergillus oryzae culture on rumenfermentation in vitro by single-factor test and orthogonal test (lq43),optimization of solid statefermentation medium and conditions of Aspergillus oryzae were determined..The results showedthat under the optimal ban10g, urea0.6%,Potassium hydrogen phosphateanhydrous0.20%,Sodium chloride0.05%,Ferrosi Sulfas0.05%and bitter salt0.05%.The optimalfermentation conditions of14%inoculation,fermentation time5d,water content of medium55%and fermentation temperature30℃.
