Abstract/Summary:
Because of beauty appearance, taste and refreshing, brightly-colored,melon is very popularin the world.Fusarium disease which affects melon of its natural growth can occur during thegrowth of the melon.Since its existence, melon production will decrease each year. Ifcircumstances is serious,it will lead to total crop failure.Not only affects the farmers’ income,but also caused some impact on the national agricultural output. Also known as Fusarium wiltdisease or Fusarium wilt, has been distributed in the world’s production of melon district, is aworldwide melons soil-borne diseases. Because fusarium wilt disease makes melon productiona serious threat, it is necessary to use scientific means to prevent this from happening.So far, thedomestic generally use pesticides to eliminate the disease to improve crop yields.May have agood effect in the short term,but disease in pesticides with a long time will have some resistanceto pesticides in the application of pesticides, it will no longer be affected by pesticides. Theexcessive use of pesticides not only pollute the environment, destroy the ecological balance, butalso brings food safety problem.Therefore, the development of biological control microbialagent requirements become very urgent.Now biocontrol has gradually entered the track, thedevelopment of biocontrol microbial agent have also made some progress, its mainly research isabout antagonistic mechanism of bacterial or fungal blight on melon. Making actinomyceteswhich is isolated from soil melon wilt on bacteriostatic experiment and Researching antagonisticbacteria culture characteristics, physiological and biochemical characteristics, growth-promotingeffect, and the impact of environmental factors on the inhibitory effect of strains.The details are as follows:(1) Actinomycetes isolated. Selected four soil samples, which came from root soil of melongreenhouses and orchards of Agricultural soil. The four areas of natural drying of soil samplesafter pretreatment with Gause’s Synthetic Agar Medium with50ug/ml Potassium dichromate forthe selective isolation of actinomycetes. A total of107strains were isolated from soils.(2) Initial screening of antagonistic actinomycetes. Determination of107strains ofFusarium antagonistic activity of the disease and make marks of them. Then keep it ready to dothe next experiment.(3) Rescreening antagonistic actinomycetes. The antagonism of all strains fermentation, thefermentation broth through a filter paper method for determination of Fusarium pathogens. Which has six actinomycetes melon wilt inhibition zone diameter≥15mm. Specifically, SY14、S14inhibition zone which can reach20mm.(4) The taxonomy of actinomyces. Combined with classical taxonomy, do some researchabout six categories of actinomycetes with its morphology, cultural characteristics, physiologicaland biochemical characteristics.(5) Antagonistic spectrum of six antagonistic actinomycetes. Six antagonisticactinomycetes conducted fungal inhibition experiments, found that six antagonisticactinomycetes also have Inhibitory effect to the other Fungi.(6) Growth-promoting effect. For six antagonistic actinomycetes experiments conductedSiderophores, phosphate solubilization experiments and indole acetic acid determination.(7) The impact of environmental factors on the activity of the strain. The six actinomyceteswere inoculated with50ml of starch in a liquid medium, depending on the fermentation time, thepercentage of carbon contained in the culture medium, PH value and the set temperature of themedium to determine different factors for strain growth activity.(8) Environmental factors affect the activity of antagonistic strains. In the role of differentenvironmental factors, six strains of what kind of change for the melon wilt antagonistic activityagainst six strains to find the best growth activity antagonistic activity and can play the bestconditions.
