Euonymus bungeanus is of Celastraceae Euonymus small tree plants, it is widely distributed in variousprovinces and regions, and it is a common courtyard ornamental tree species. Researchers have found that itsseeds, roots, bark have rich economic and medicinal value. We selected Euonymus bungeanus fruit for thisstudy, determination of the content of its nutrients and conducted researches on its alkaloids and saponinsextraction process, isolated and purified the saponin, then conducted preliminary studies in antibacterial andanti-tumor cell activity of saponin components.(1) To analysis the nutritive elements of potassium, calcium, magnesium, sodium copper, iron, zinc andso on in Euonymus bungeanus, the result showed that: There are rich trace elements in Euonymus bungeanus,such as calcium, magnesium, sodium, potassium. The total mass fraction of amino acids to reach16.372%,inwhich the mass fraction of the essential amino acids average of5.694%(2) In the study of the extraction process of Euonymus bungeanus fruit alkaloids, we took hot waterextraction method and ultrasonic extraction method to extract alkaloids from Euonymus bungeanus fruitrespectively, optimized the extraction conditions by orthogonal experiment, and determined the optimumextraction process of Euonymus bungeanus fruit alkaloids were: extraction temperature of80℃, ethanolconcentration of80%, extraction time of1.5h, solid-liquid ratio of1:25and extracted three times, under theseconditions the alkaloid extraction ratio was22.6%; the optimal conditions of ultrasonic extraction methodEuonymus fruit alkaloids are: Extraction temperature of70℃, ethanol concentration of80%, extraction timeof50min, solid-liquid ratio1:20and extracted three times, the alkaloid extraction ratio was24.1%.(3) In the study of the extraction process Euonymus bungeanus fruit saponin, we took hot waterextraction and ultrasonic extraction method, then it was optimized by orthogonal experiment to get the bestcondition of the extraction process were: extraction temperature70℃,60%ethanol, extraction time2h,solid-liquid ratio of1:15. And the saponin extraction rate could get to58.7%under those conditions byextracting two times; the best conditions for the ultrasonic extraction method were: extraction temperature70℃,70%ethanol, extraction time50min, solid-liquid ratio1:15. The saponin extraction rate could get to75.3%by extracting three times under those conditions.(4) The next experiment was taken using silica gel column chromatography for separation andpurification of saponin. In this step,10%~90%ethanol was used to elute saponin, finally two saponinfractions of Euonymus bungeanus fruit were got: Ebs-a, Ebs-b. The Ebs-a, Ebs-b yields were43.8%,24.3%,respectively, through D152macroporous resin column. Also, Ebd-A, Ebs-B were only got two singlesymmetrical peak which were eluted by D152macroporous resin column, which showed that those two saponin components all had both a certain uniformity.(5) Also, the experiment of antibacterial activity for Euonymus bungeanus on peach fruit saponins wasconducted, which studied the cytotoxicity of saponin components for Bacillus subtilis, Staphylococcus aureus,Escherichia coli, Salmonella. The results showed that the effects of Euonymus fruit saponins on four kinds ofbacteria were all inhibited, especially for Galanz negative bacteria Bacillus subtilis and Salmonella.(6) Finally, the experiment for on anti-tumor cell activity using Euonymus fruit saponins was carried outto study the cytotoxicity of saponins on human hepatoma cell line SMMC, human cervical cancer cell lineHeLa, human gastric cancer cell line BGC823, breast cancer cell line MCF7, canine kidney cell line DK. Theresults showed that there was a significant inhibitory effect of Euonymus fruit saponins on the growth ofhuman hepatoma cell line SMMC, human HeLa cells and breast cancer cell line MCF7, no significantinhibitory effect on human gastric cancer cell lines BGC823, and almost no any damage on canine kidney cellline DK with a normal cell growth.
