Koi herpesvirus (KHV) was identified as pathogens cause high mortality in koijuvenile and adult in1998, the first outbreak has been reported in Magan MichaelIsrael and the U.S. The virus has spread to most continents so far, Outbreaks havebeen reported in Belgium,South Africa, France, Denmark, Austria, Italy,Luxembourg, Poland, the Czech Republic, Thailand, Japan, Malaysia, China andTaiwan and South Korea. The death of koi/carp which cause by KHV bring hugeeconomic losses,and the desth of native carp cause a lot of omnivorous fish disappearfrom freshwater ecosystems, which may alter the population structure and ecosystemfunctions.Our study chosen two outer membrane fragments of ORF68gene that genotypesin stable of the three KHV. Recombinant plasmid of the two fragments was built byprokaryotic expression,and the753bp fragment was successed.Immunization andconteracting toxic substances to reseach the purified protein. The results showed thatthe immune carp antibody levels is the highest in28th days, and after theconteracting toxic test the protection rate is28%.
