This paper systematically studies the combined effects of dietary roughage of donkey and the impact onthe microbial flora of the cecum. The experiment consists of three experiments:In the first experiment, the study investigates the effects of different coarse material combinations on invitro donkey cecum. In the experiment, a systematic design method was applied, and concentrate androughage ratio was4:6. Coarse material combinations were corn stalks and alfalfa(3:7,5:5,7:3), straw andalfalfa(3:7,5:5,7:3), corn stalks and straw(3:7,5:5,7:3). In the artificial environment of cecum, PH, NH3-N,VFA, degradation rate of DM and CF fermentation would be measured after24hours in vitro. Meanwhile, bymaking the PH, NH3-N, VFA, degradation rate of DM and CF fermentation as evaluation factors, theexperiment evaluated nine different combinations of diets. The experimental results show that under differentcombinations of coarse material structure, the maximum associative effect was shown when the ratio of Cornstalks and alfalfa is5:5.In the second experiment, the study investigates the effects of different coarse material combinations ondonkey cecum microbial flora. The experiment used phenol extracted DNA of Cecum microbial on which theUV spectrophotometer and the gel electrophoresis were performed and the results showed the DNAconcentration was between219and290ng/ul, and most of the DNA chains were above15,000bp withcomplete structure. Entodinium and3important fiber degradation bacteri in the cecum of donkey content ofthe Nine groups were quantified through SYBR Green fluorescent quantitation PCR. The results show that thecomposition of Roughage diet has a significant impact on the main cellulolytic bacteria structure in donkeycecum,With varying degrees of impact on different cellulolytic bacteria. There are obvious changes in theamount of the number of Ruminococcus albus.The chang of entodinuim is the smallest.
