Trichokonins is a extraction from the green trichoderma viride, It is a kind ofnew plant growth regulator, in the study of it is to be in the ascendant. This paperexplored the effect of Trichokonins to seed germination; We measured the changeof the seeds main nutrients and activities of protective enzyme in the soaking and the process of seed germination under different concentration of Trichokonins. Withwater and Naphthalene acetic acid (NAA)for comparison, We further studied Sophora japonica seedlings on the root-shoot ratio, plant height, leaf area and accumulation law of various kinds of organic matter content in the leaves, and of all kinds ofchanges on the activity of protective enzyme of the leaves under Trichokonins different treatment and different sampling time; We scanned and analyzed Sophora japonica seedling root by WinRHIZO root analysis system, After the treatment of Trichokonins,We explored the basic forms of root system(including root length, root area,root volume, root tip count, etc.) and Link-topology-development status, etc.ofSophora japonica seedlings. And we explored the the root activity and all kinds of protective enzyme activity changes of the root.1. The effects of Trichoderma koningii on Sophora japonica seeds’germination ofphysiological and biochemical indicatorsWe concluded that Trichokonins in the concentration of0.3mg/L, it can improve seed germination rate, germination potential, germination index, it accelerate theradicle and embryo length of Sophora japonica, it reduce the electrical conductivityof Sophora japonica, decrease the loss of nutrients, it increased all kinds of protective enzyme activity in the seed. Research finding, In the concentration0.3mg/LofTrichokonins, It is good for Sophora japonica seed germination, by this time, Sophora japonica seed germination rate, germination potential, germination index was respectively higher than that of control29.33%,24.66%,3.20, the radicle and embryo was respectively longer than the control by11.72mm and5.37mm. By comparing, the electrical conductivity is lowest in concentration0.3mg/L of Trichokonins seeds soaking with24h, the reduce degree was the lowest, It could protect the inte grity of the cell membrane; by this time, It reduced the loss of nutrients of solubleprotein, soluble sugar, starch, persistent most of the organic matter, respectively38.15%,43.47%,32.46%higher than that of control. To a certain concentration extent,it increased all kinds of protective enzyme activity in the seed by the treatment ofTrichokonins, Compared with the control, increased by122.36%and49.05%respectively.2. The effect of Trichokonins on physiological and biochemical indicators in Sophorajaponica seedlingsBy comparing different Trichokonins concentrations which filling root and sprayingto Sophora japonica seedlings, We found that it had significant promoting effect ofTrichokonins on0.3,3mg/L for Sophora japonica seedling. When filling root processingof Trichokonins was in the concentration of3mg/L, The root-shoot ratio of Sophorajaponica seedlings increased by41.67%, plant height, leaf area, respectively increased by75.99mm,51.07cm2compared with the control. when spraying processing ofTrichokonins was in the concentration of0.3mg/L, The root-shoot ratio of Sophorajaponica seedlings increased by39.16%, plant height, leaf area, respectively increased by71.04mm,81.86cm2compared with the control. When filling root and spraying ofTrichokonins was in the concentration of0.3mg/L, spraying of naphthalene acetic acidwas in the concentration of40mg/L, the soluble protein, soluble sugar and starch contentof Sophora japonica seedlings were in the most. And Trichokonins spraying is better thanthat of naphthalene acetic acid, respectively increased by70.18%,82.62%and142.14%compared with the controls. In addition, after processing, it improved all kinds ofprotective enzyme activity in Sophora japonica seedlings leaf with filling root andspraying of Trichokonins in the certain concentration range. Compared with naphthaleneacetic acid spraying processing, The effect of Trichokonins spraying treatment is good. Theresults show that the experiment reduced all kinds of material contentthe and activity ofprotective enzyme of Sophora japonica in300mg/L high concentrations of Trichokonins.Ithas inhibitory effect on Sophora japonica seedlings.3. The influence about Trichokonins to Sophora japonica seedling root growth We scanned and analyzed Sophora japonica seedling root by using WinRHIZO rootanalysis system, the results showed that When spraying processing of Trichokonins was inthe concentration of0.3mg/L, the total root length, total projection area, total surface area,total volume, root number of Sophora japonica seedling were the maximum, respectivelyincreased by84.41%,204.05%,279.54%,51.96%and136.15%compared with thecontrols. Compared to300mg/L concentrations of Trichokonins reduced the total rootlength, total projection area, total surface area, etc.of Sophora japonica. When sprayingprocessing of Naphthalene acetic acid was in the concentration of20mg/L, the total rootlength, total projection area, total surface area, total volume, root numberof Sophorajaponica were largest, respectively increased by54.25%,186.87%,181.53%,26.54%and63.38%compared with the controls. less than0.3mg/L Trichokonins spraying process,significant difference. That Trichokonins in promoting root growth and developmentaspects of the effect is better than naphthalene acetic acid. Compared to300mg/Lconcentrations of Trichokonins reduced the total root length, total surface area, etc.ofSophora japonica. The growth of root system of the Link topology-reflects the root,Sophora japonica seedling root Link was increased, and the path of peripheral populations,height, root number increased, It further proved that the extension of Sophora japonicaseedling root and lateral root growth. By paraffin section shows that Sophora japonica rootroot tip meristematic zones cell populations, increased cell elongation elongation zone afterTrichokonins treatment compared with water control.
