The height control of ornamental plant can be realized the purpose of increasing ornamental qulity, controlling the plant height, fostering the new type of dwarfed flowers, will greatly expand the flower industry development space.There are many genes that regulate the plant height, many of them affect gibberellic acid synthesis. Gibberellic acid is the most important growth hormone that can control the plant height, GA2-oxidase is the key enzyme that encoded gibberellic acid synthesis, catalyzed the bio active GA change for the activity of GA. GA2-oxidase overexpression, would reduce the level of endogenous GA, so as to achieve the purpose of making plant dwarfed. GA2-oxidase gene not only promote the stem elongation and growth, also influence flowering and fructification.The target gene expressed in the plant successful can reduce plant height,and make flowering and fructification abnoramal. Choosing specific promoter is a better choice.In this experiment we cloned arabidopsis stem specifically expressed promoter, obtained the arabidopsis GA2-oxidase gene, constructed GA2-oxidase gene overexpression vector and specifical expression promoter.We introduced the gene into petunias via agrobacterium-mediated technique, obtained dwarfed transgenic plants. Using specific promoter regulate GA2-oxidase gene expressed in plant specific organ, making gibberellic acid level changes restricted in the stem, overcoming the problems of flowering and fructification because of gibberellic acid"s pleiotropic fostering the new type of dwarfed flowers.In this study we cloned the gene of AtGA2oxl. constructed the AtGA2oxl overexpressed carner; Isolated the promoter sequence of and At3g56700gene, constructed the specifical expression vector of AtGA2oxl gene. Introduced the gene into petunias via agrobacterium-mediated method, observing specific promoter’s actual effect.This experiment’s main works and results are as follows: 1. The gene of AtGA2oxl was cloned, the sequence’s length is1224bp.2. Construction of AtGA2oxl overexpressed carrier pCMF501, introduced the AtGA2oxl gene into petunias,14independent Kan-resistent plant were obtained. Observe the effect of AtGA2oxl gene for plants in the future time.3. The promoter of gene A t3g56700is cloned, the promoter size is2147bp.4. Construction of specifical expression vector pCMF712fused by AtGA2oxl gene and promoter of At3g56700gene.
