The early pregnancy factor (EPF) is a pregnancy protein, EPF is great significance that predicted early embryonic death and abortion, identification of embryo transfer, monitoring the status of the fetus as well as the importance of the diagnosis of esophageal cancer. EPF is an immunosuppressive protein detected in the serum in early pregnancy.The research contents of this study were as below. Comparing the blood of 4 non-pregnant cattle with 12 cattle which have familiar physical and feeding state but different gestational period(5 for 1-2 months; 4 for 3-5 months; 2 for 6-9months), and detect activation of EPF in the blood through rosette inhibition test (RIT). Based on the analysis of titer for rosette formation, in order to ascertain the time when the best activity and largest concentration can be detected during the pregnancy period, which can be facilitate the larger scale extract of EPF. Obtain EPF through DEAE-Sepharos F.F and CM–Sepharose F. F ion-exchange column chromatography and Con A-Sepharose 4B and Heparin Sepharose Cl 6B affinity chromatography from healthy calver serum when EPF was secreted at the largest concentration. Concentration and molecular weight of EPF were measured by staining of Coomassie brilliant blue R250 and SDS-PAGE respectively, which turned out a protein band at 53.6 Ku..It is prepare to further development early pregnancy factor monoclonal antibody Colloidal gold test strip of livestock abortion and infertility.
