| This paper used a high sensitivity of immunohistochemistry streptomycin avidin - peroxidase method (SP) and in situ end labeling (TUNEL) method, combined with computer image analysis to study the expression and distribution of apoptotic proteins Caspase-3, Caspase-9, Bcl-2 and Bax during different duration (early, middle and late) of goats pregnancy,and maped out the curves of relative expression of apoptotic proteins and CL apoptosis respectively, which could reflect the relationship between the expression of the four above-metioned apoptotic proteins and the luteal apoptosis directly , and meaningfully,to provides a theoretical basis for the studying of the physiological characteristics of corpus luteum. Some results was achieved as follows:1. Early period of pregnancy: the expression of Caspase-3, Caspase-9 and CL apoptosis are decreased parallel, while the Bcl-2 and Bax are increased slightly.2.Trimester: the trend of luteal apoptosis increased continuously and Caspase -3 and Caspase-9 rised up at first and then decreased, and reached the highest value on 40d; Bcl-2 and Bax were tested decreased and Bcl-2 declined more than Bax.3.Late pregnancy: both of CL apoptosis and Bax expression are decreased, whereas Caspase-9 and Bcl-2 are slowly rising, and Caspase-3 was increased sharply.These results suggest that: Bax was positively correlated with luteal apoptosis and negatively correlated with Caspase-3 and Bcl-2 respectively,but nothing with Caspase-9;Caspase-3 and Caspase-9 was significantly correlated with each other. The overall relationship between four apoptotic proteins with respect to relative positive expression was showed as lower during early pregnancy, higher during late pregnancy and the the changes during the mid pregnancy were variable. |
