Taraxacum Kok-saghyz Rodin is herbaceousperennid of the family Compositae. It is one of the world’s major rubber yielding plants. The quality of the natural latex from TKS is similar to the rubber tree’s, both of which can be applyed to the rubber industry.At present, the study of TKS latex protein is rarely reported at both home and abroad. ’This study was conducted to establish a two-dimensional electrophoresis(2-DE) method of proteins from TKS latex by using proteomics. After preliminary analysis of TKS latex by protein abundance, proteins related to the synthesis of rubber latex were found out, we also provide a new angle for the research on the mechanism of the rubber latex biosynthesis. The results obtained are as follows:1. The experimental results demonstrate that Tris-saturated phenol extraction could be a considerable method for TKS latex protein extraction. Extracted proteins that are suitable for 2-DE, are the foundation to obtain precise 2-DE map.2. High quality 2-DE map were obtained by optimizing 2-DE conditions(including IPG strip, protein loading quantity and different ways of protein staining).3. With protein spectrum,11 proteins having a positive result of the experiment were identified, a chunk of bio-active proteins were obtained after database searching.Including Resistance stress related (ascorbate peroxidase, chloroplast polyphenol oxidase)、The cytoplasm skeleton changing (actin) and glycolysis involving proteins (Triosephosphate isomerase)4. According the functional analysis,we foud out enolase and ascorbate peroxidase both exist in the latex of Rubber tree and TKS.
