Powdery mildew disease is the main cause of wheat yield decreased worldwide. In the previous study conducted by our laboratory, TaWRKY34 gene was identified in response to wheat powdery mildew fungus by cDNA macroarray and semi-quantitative RT-PCR. We now conducted a study to investigate the role of TaWRKY34 in Powdery mildew resistance by Virus-induced gene silencing (VIGS) and transient over-expression. We aim to get the over-expressing and knockdown of TaWRKY34 in To generation and conduct primary analysis of gene function.This will provide substantial basis for Transgene.We predicted the conserved domain of WRKY by comparing protein sequences, got the siRNA targeted sequences of TaWRKY34 by siRNA Target Finder. Then we silenced the expression of TaWRKY34 in near-isogenic line of Pm16: Pm16/beijing837 and analyzed its function in Powdery mildew resistance. The result showed that the expression of TaWRKY34 was down-regulated in VIGS-treated plants which displayed enhanced resistance to Powdery mildew (Occurrence of disease symptom was delayed) and suggested that TaWRKY34 may be a negative regulator in Powdery mildew resistance of Pmi 6/beijing837.Furthermore, we constructed the binary vector for over-expression of TaWRKY34 in wheat leaves through microinjection bombardment. Then the Powdery mildew resistance was evaluated in epidemic cells expressing TaWRKY34 transiently.The result showed that over-expression of TaWRKY34 might be a negative regulator in Powdery mildew resistance of Pm16/beijing837.
