African daisy (Osteospermum hybrida) that is from compositae ostespermum, is a new ground-cover and potted flower with high flower dense and flamboyant color. It is of great ornamental and economic interest. Presently, the lack of study of african daisy, and the seeds have to import mostly, so the cost is high. Because the heat and cold hardiness is not so good, african daisy is limited in virescence uitiliztion and popularization. So african daisy s introduction, improvment, new germplasm selection and breeding, is mostly important now.In the present paper, traditional morphorlogical method and hundred-mark methord were used to select superior individuals. Regeneration system was estanblished by tissue cultue. Based on the pre-reseach, molecular mark technology was employed to validate Somaclonal Variation, and germplasm innovation of Osteospermum hybrida was studied. The purposes of this research is to provide a basis of theory and technology for african daisy s breeding, popularization, and the use of Somaclonal Variation to select and breed new cultivars, produce new germplasm.1) seedlings propagationBased on the biological characters of african daisy that is from Flower Reseach Institute of Italian (San Remo), the suitable seeding time in WUHAN is in autumn (the average temperature is about 15℃).2) Superior individuals selection by hundred-mark methordBased on the african daisy’s characteristics and 50 professional and laypeople preference investigation result of flower color and ray floret type, the evaluation standard of flower color and ray floret type is estanblished. And the evaluation system and standard of african daisy was estanblished by hundred-mark method.14 superior individuals that was suitable in WUHAN with high ornamental value was selected by evaluation system and standard.3) Rapid propagation in vitro system was estanblishedInduction of shoot cluster through axillary bud sprouted capacity from stem explants of superior individual’KP002’was studied. And the rapid propagation in vitro system was estanblished.The optimum medium of shoot cluster and adventitious induction is MS+6-BA0.2 mg/L+KT0.5 mg/L.4) Leaf regeneration system was optimizedLeaf regeneration system was optimized in the present paper. The suitable medium for the callus induction and subculture from leaf explants of superior individual’KP002’ is MS+6-BA1.0mg/L+NAA 0.75mg/L. The subculture medium adding 2g/L acticarbon exacerbated callus browning, and was not good to callus growth.Then the callus was transferred on the medium (MS+6-BA1.0 mg/L+NAA0.1 mg/L), which got the regeneration of shoots. And the plants has the fine rooting capacity on 1/2MS+NAA0.5 mg/L medium. There are great difference between individuals and genes. And the influence of different individuals or genes in tuissue cluture, espically in shoot cluster induction need further study.5) Somaclonal variation assay by ISSR makerInter simple sequence repeat (ISSR) marker assay was employed to validate Somaclonal Variation of Osteospermum hybrida, and the result showed that the variation rate of callus was not correlated with the number of successive culture, and the variation rate of the plantlets, regenerated by 2nd successive culture of callus reached to 30.23%, compare with maternal parent. Somaclonal, from leaf regeneration system existed high variation that can be used for Osteospermum hybrida new cultivars breeding and seletion.
