In this thesis, a study has been made on physiological status of Chlamydomonas reinhardtii when subjected to various deficiencies of nutrient elements, such as nitrogen, sulfur, phosphorus, and combination of these elements. The research also attempted to investigate the lipid accumulation and fatty acid side chains of lipid and hope to find out the regulations during accumulation.Based on the premise that Chlamydomonas can accumulate lipids when suffering deficiency of nitrogen, the study inspected physiological status of cells under combination deficiency with nitrogen, including cell numbers, sizes, and weights. When subjected to nitrogen deficiency, cells stopped dividing and started to accumulate lipids, being obese. When nitrogen deficiency is coupled with sulfur deficiency, it didn’t come with acceleration or inhibition to cell obesity. When nitrogen deficiency is coupled with phosphorus deficiency, it showed inhibition to cell obesity. When subjected to sulfur deficiency, cells could develop as usual for some time but began to shrink when subjected to phosphorus deficiency.Then, the dissertation probed the cell disruption and found out that high-pressure breaking have more advantages than other methods. When extracting oil from disrupted cells, ultracentrifugation can avoid chlorophyll interference caused by Soxhlet extraction or chloroform- methanol method effectively.For the reason that the research had high acidity, the lipid dissertation compare different kind and dosage of catalyst for interesterification. The dissertation concluded acid-catalysis was more appropriate for the lipid in Chlamydomonas and analyzed the side chains of the lipid. The study found the chains changed when deficiency changed.
