| We mainly focused on the treatment method of controlling the M.aeruginosa.We screened the effective inhibition of M.aeruginosa materials and determined the effective concentration.According to the screening test results and actual using requirements,we choose several materials to optimize the algaecide agent,which provides reference and guiding significance to the research and development of drugs of controlling algae.1.Screening of algae inhibiting materias and determination of the optimal inhibiting concentration of M.aeruginosaIt was aimed to screen effective materials for inhibiting M.aeruginosa and to determine the optimal inhibition concentration.In this experiment,one of the algae biomass namely Chlorophyll a detection method was used to determine the biomass of M.aeruginosa,to study the inhibiting effects of algae inhibiting materials on M.aeruginosa,and two times dilution method was used to determine the best inhibitory concentration of 9 kinds of drug materials.Result:Radix et Rhizoma Rhei,Rhizoma Coptidis,Cortex Pseudolaricis,Tangerine peel, Ma Shui orange peel,Pomelo peel,TieGuanyin tea Allium sativum and salicylic acid can effectively inhibit the growth of M.aeruginosa,while Hordeumvulga and Herba Agrimoniae can not.When the extract of Tangerine peel,Pomelo peel and Ma Shui orange peel and salicylic acid and TieGuanyin tea,Radix et Rhizoma Rhei and Cortex Pseudolaricis,Rhizoma Coptidis were 6.25g/L,3.13g/L,128mg/L,5g/L,2.5g/L,it had a significant inhibition effect on growth of M.aeruginosa,and the effect of inhibiting algae increased with the increase of the concentration of each extract.Considering every angle it was more reasonable to set the above control algae concentration.It was the first time that screened algae inhibiting materials have been determined the optimal inhibition concentration,which is beneficial to understand the research of controlling cyanobacteria and study further.2.The study on the mechanism of inhibiting the M.aeruginosa by materialsWe selected citrus peel,salicylic acid and garlic from the nine effectively inhibit algal materials in the first chapter, and observed the effect of the three kinds of materials on the growth of M.aeruginosa physiological biochemical several indexes. Results showed that in the fourth days three different kinds of citrus peel of verdigris microcystin algae growth inhibition rate were 85.83%,90.55% and 90.55% respectively under the normal temperature processing.The inhibition rate were 71%,81%,77.42% under high temperature treatment.The seventh day the inhibition rate was more than 94.38%.The content of chlorophyll a in 0.12g/L treated group was lower than that the control group in 12 h,and the difference was significant(P<0.01),and the content of PC was too little and can not be detected in 96 h. The content of APC had a growth trend.Content of PE maintain at the initial level.Compared with the control group, the total content of soluble protein in experimental group decreased 33.3% in 24 h and only 2.2% of that in the control group in 96 h.There was a downward tendency of the content of PC in group of 6.4 g/L of Garlic.The content of APC changed quite gentle before 24 h.The content of PE changed little.The content of total soluble proteinin was at 45.0% of the matched group in 48 h,as low as 21.3% in 96 h.This study proved that the citrus contained algal inhibiting components,and can keep activity under high temperature treatment.The content of Chlorophyll a,algal bile protein and the soluble protein can be influenced by salicylic acid and garlic.The salicylic acid and garlic controled the growth of verdigris microcystin algae by photosynthetic system â…¡ of the algae.3.The study of optimization and stability on the algae inhibiting agentAccording to the previous research, the orthogonal experiment was carried out on the three materials in the second chapter to obtain the optimal combination of the algae inhibiting agents, and the stability on the algae inhibiting agent were studied.The results showed that the optimal combination of the rectangular test method was A2B3C1,namely algicide the optimum formulation is 6.25g/L of tangerine peel extracts,256mg/L of SYS,0.625g/L of garlic,and SYS plays a leading role in the content of chlorophyll a on M. aeruginosa.There was no significant difference in the inhibitory effect under normal temperature and boiling water heating 20 min, 40 min and 60 min.When the algaecide agent was stored at 4 degrees,it can maintain a good inhibitory activity, and at room temperature was slightly,and worst at-20. In experiments with different inoculation,the algaecide agent can effectively inhibit M.Aeruginosa,which OD680 nm is below 0.4,and it can influenced the composition of phycobilisome protein,PC content in each treatment group showed decreasing trend, APC was slightly increased,while PE remained basically unchanged,total soluble protein content reduced.The optimization ofalgicidal agent can effectively inhibit M.aeruginosa concentration,and algae photosynthetic system II is a site of action of the algaecide agent for growth and reproduction of M.aeruginosa. |
