| Sunflower is planted widely in China, the fruits of it are sunflower seeds, which contains a certain amount of chlorogenic acid, and sunflower seeds compared with other sources such as honeysuckle and eucommia bark, sunflower seeds are in low price.However, the study on the chlorogenic acid in sunflower seeds is relatively few in our country. Chlorogenic acid itself is known as the "plant gold", it not only has the function of antioxidation, antitumor, but also has the physiological characteristics which possessed of effective antilipemic activity and reducing the blood pressure. The chlorogenic acid in sunflower seeds can be oxidized easily into other substances, which has an adverse effect on the subsequent protein extraction, colour and lustre and nutritional value. Therefore, the yield of chlorogenic acid from sunflower seeds has important economic value. The sunflower meal after the extraction of chlorogenic acid as feed usually, the protein resources are wasted, thus we aimed to use the sunflower meal as raw material to make protein beverage, which can improve the comprehensive utilization of the added value of the sunflower seeds.Taking sunflower seeds as raw material in this test, the first research is the extraction of chlorogenic acid from sunflower seeds by the method of traditional ethanol,then the next step is the extraction of chlorogenic acid from sunflower seeds by the ultrasonic and microwave method assisted, and optimize the process, and then select the macroporous resin purification of chlorogenic acid, and the features of chlorogenic acid is studied, finally the sunflower meal after the extraction of chlorogenic acid as raw material, the preparation of protein milk beverage. The results are as follows:(1) The research on the ethanol volume fraction, liquid ratio, power of ultrasonic wave, microwave power and microwave irradiation time on the extraction yield of chlorogenic acid in single factor experiment is respectively studied, and then according to the Box- Behnken response surface design of experiment selected three factors which have large effect, the liquid ratio, ultrasonic power and microwave power are optimizated, the best reaction conditions is obtained which the liquid ratio of 25:1(mL/g), ultrasonic power 300 W, microwave power 540 W. Three repeating validation tests is operated under the optimal conditions, the extraction yield of chlorogenic acid is 3.27%.(2) The resin of NKA-2 is selected for purity the chlorogenic acid crude extract, and determine the sample concentration of 6 mg/mL, the sample flow rate of 2 mL/min, eluent concentration of ethanol was 90% finally, under the condition of the purification, the purity of chlorogenic acid is 61.92%.(3) To study the features of chlorogenic acid extract, the hydroxyl free radicals, DPPH and superoxide anion free radical is showed crespectively, which compared with the scavenging capacity of Vc, when the concentration of sample solution is 60 μg/mL, the hydroxy free radical clearance rate reached 66.53%, the DPPH free radical clearance rate reached 80.17%, the ultra oxygen anion removal ability is 65.04%, the results showed that the chlorogenic acid is a good way to instead of Vc.(4) The outgrowth of the sunflower meal after the extraction of chlorogenic acid as raw material in the study, the preparation technology of sunflower protein beverage is studied, finally we determined the best ratio for preparation of sunflower protein beverage that is the granulated sugar addition amount is 9%, citric acid content is 0.2%, the adding quantity of milk powder is 3.5%, the addition amount of composite additive is 0.2%. Verification experiment was performed under the optimum conditions, the sensory score is measured as 92 points. |
