Object: Define the preparation technology of Baixuanxiatare Ointment, establish assay method of the content of Quercetin and Gallic acid and investigate its stability.Methods:(1)Extraction technology: research extraction technology of Baixuanxiatare Ointment by orthogonal experiment; Optimize extraction technological parameter of medicine by using the quality of extract and the content of Quercetin and Gallic acid as index;(2) Forming technology; Use AGAR gel diffusion test to determine the type of ointment; Optimize the formula by applying orthogonal experiment and using the smooth level, coating ability, moisture retention and cleanability as index;(3)Assaying: HPLC was applied in assaying the content of Quercetin and Gallic acid of Ointment(4) Study the preliminary stability of the ointment produced by above methodResults:(1) The optimum extraction technology is mixing humifuse euphorbia herb and 70% ethyl alcohol at 1:8, heating reflux and extracting 3 times, which takes1.5 hour for each time. Combine filtrate after filter, decompress and concentrate it. Mix dreg of humifuse euphorbia herb with Terminalia Chebula Retz, add ten times water and decoct it 3 times, which takes 1.5 hour for each time. Combine filtrate after filter, decompress and concentrate it. And then mix it with humifuse euphorbia herb alcohol extraction and concentrate the mixture by relative density 1.2~1.3.(2) The best prescription is that extract the octadecyl alcohol 3g, white vaseline 12 g, liquid paraffin 5g, poly isosorbide mononitrate 80.6g, 2g of lanolin, glycerin 12 g, Aloe powder 28 g, Scammoniae resina 10 g and add water up to 150g;(3) Quercetin content determination method: Mobile phase: methyl alcohol – 0.4% phosphoric acid liquor=50:50(V/V); Column temperature: 25℃; Flow rate: 1ml/min, wave length: 370 rm, standard curve: y=2.1×107x+5.5×105,r=0.9997,n=6. Gallic acid content determination method: Mobile phase: methyl alcohol – o,1% phosphoric acid liquor=10:90(V/V); Column temperature: 25℃; Flow rate: 1ml/min, wave length: 270 nm, standard curve: y=8.4×106x+4.5×105,r=0.9995,n=6;(4)Ointment appearance is not changed under the low temperature and high humidity and the content of Quercetin and Gallic acid is stable; Character will be changed when it’s under high-light; Its content will bereduced by high temperature.Conclusion: The extraction technology of Baixuanxiatare Ointment is stable, reasonable and doable; water-in-oil Ointment produced by optimized prescription has stable properties, appearance of bright color, fine texture and high moisture retention. And it has high coating ability and cleanability; The determination method is simple and accurate; The Ointment is not sensitive to strong light and high humidity and its content is stable under the airtight preservation at room temperature...
