Trichosanthes kirilowii seed contains about 26.75%~28.82% protein. Study showed that trichokirin and glycoprotein of trichosanthes kirilowii seed proteins have been used in the treatment of diseases such as cancer. This article adopted the fat-free trichosanthes kirilowii seed powder as raw material, studied several extracting methods of trichosanthes kirilowii seed protein, and the characteristics of trichosanthes kirilowii seed protein isolate were also researched. Through the research, provided basis for comprehensive and effecient utilization of trichosanthes kirilowii seed protein in future. Meanwhile, layed the foundation for the deep processing of trichosanthes kirilowii seed, and from food to medicine, health care products in future.This paper studied the alkali soluble and acid sinking method of extraction of trichosanthes kirilowii seed protein, and this method has high extraction rate, low cost etc.Using the single factor test and orthogonal test to determine the optimum process of trichosanthes kirilowii seed protein.Under the fixed condition of size 100 mesh, acid sinking pH 3.8, optimum process conditions of extracting was:temperature of 70℃, alkali soluble pH 10.0, solid-liquid ratio 1:20,80 min of time. Trichosanthes kirilowii seed protein extraction yield could reach 71.10%,purity was 77.01%.Enzymatic method had much advantages such as mild reaction, good specificity and less environmental pollution, etc. Due to the specificity of the enzyme, the conditions and results of reaction between different enzyme and its substrate were not identical. This paper compared the extracting effect of several kinds of enzymes of trichosanthes kirilowii seed protein. The tests showed that the extraction efficiency of alkaline protease was 60.48%, and much higher than other several kinds of enzymes, so we adopted alcalase for using enzymes. By single factor experiment and response surface analysis method, in fixing time 150min、acid sinking pH 3.8, and then obtain the best extraction process parameters for:solid-liquid ratio 1:30.4. plus enzyme amount 3.0%, pH value of 9.9, temperature 64.8℃. The enzymatic extraction yield was 73.54%, purity was 84.78%.The paper compared 2 kinds of purification methods. Determined the amino acid composition and content of trichosanthes kirilowii seed protein. Its amino acid composition was entirety, essential amino acids composition fitted the human body needs. Test also analyzed the solubility, water and oil absorption, emulsification, emulsifying stability, foaming ability and foam stability of trichosanthes kirilowii seed protein. It showed that trichosanthes kirilowii seed protein has good solubility. Compared with soybean protein trichosanthes kirilowii seed protein was better in water absorption, emulsification, emulsifying stability. And it’s oil absorption, foaming ability and foam stability were lower than soybean, but it still had good processing properties.
