The research studied the optimization of extraction the oil conditions, used GC-MS to analyse the chemical compositions, and also studied the scavenge free radicals, antibacterial effect, afford theoretical basis for depth development Chrysanthemum morifolium, in order to enhance the economic value. The main results as follows:1. Optimization of extraction process for Chrysanthemum morifolium essential oil.Combined with soxhlet extraction and steam distillation, Chrysanthemum morifolium essential oil in two-step. The effects of particle size, ultrasonic time, solid-to-liquid ratio and distillation time on the extraction rate of Chrysanthemum morifolium essential oil were investigated by one-factor-at-a-time experiments, used four factors and three levels of response surface methodology to optimize of extraction process for the essential oil. The results indicated that the optimal conditions were particle size 80~100 mesh powder, ultrasonic time 30 min, solid-to-liquid ratio 1:11, and distillation time 9 h. Under these conditions, the extraction rate of Chrysanthemum morifolium Essential Oil was 4.80 mL/Kg.2. The determination of the Chrysanthemum morifolium essential oil composition and content. GS-MC was used to analyse the the chemical compositions of Chrysanthemum morifolium essential oil. The results showed that 61 chemical compositions were isolated, the content of haplotype terpenes and sesquiterpenes were 20.964%; the content of esters were 2.03%; The content of alcohols, phenols, aldehydes, ketones and organic acids were 6.73%; The content of olefin were 19.26%; The content of alkanes were 11.28%.3. Antioxidant capacity of research of the Chrysanthemum morifolium essential oil. Ability to eliminate free radicals in vitro, are used to indicate the Chrysanthemum morifolium essential oil antioxidant capacity. The results showed that 10% of the oil solution of DPPH·clearance rate was as high as 69.88%, clearance rate of OH was 51.84%, clearance rate of O2-· only was 40.42%, but its ability to remove free radicals were less than 2% of PG. Under the same concentration, the free radicals removal ability order was DPPH>OH>O2-·, the oil can scavenge free radicals, and with the oil increase the ability stronger of scavenge free radicals.4. The antibacterial ability of research of the Chrysanthemum morifolium essential oil. Studied the antibacterial activity of Chrysanthemum morifolium essential oil in vitro, and explored the influence of surfactant, temperature, pH and UV irradiation for antibacterial stability, the results showed that the oil all had certain antibacterial activity on the microorganisms, such as Escherichia coli, Staphylococcus aureus, salmonella, pseudomonas aeruginosa and Bacillus subtilis. The minimum bacteriostatic concentrations of the oil for various microbes were as follows: Escherichia coli and Staphylococcus aureus 1.30%; salmonella and Bacillus subtilis 0.67%; pseudomonas aeruginosa 0.325%. The influence of surfactant on the antibacterial stability was obvious, but had better stability of temperature, pH and UV irradiation.
