The fruiting body of Gloeostereum incarnatum is rich of nutrient, and its polysaccharides have the physiological function of broad-spectrum antibacterial characters, enhancing immune activity and inhibiting tumor cell. They have been extensive applied in the preservation of food, pharmaceutical and clinical areas, and green feed additives. The research of medium composition on submerged fermentation and polysaccharides extraction of Gloeostereum incarnatum are infrequent and still in the initial stage of exploration. The culture conditions of liquid fermentation of mycelium and extraction of polysaccharide were studied in this article, and the mycelium polysaccharide purification was also discussed. These results can provide a reliable basis on making a class of drugs for the treatment of acute enteritis and ulcerative colitis.In the study of culture conditions study, Different carbon source, nitrogen source, and growth factors were selected on elm fermentation biomass.The amount of maltose^ soybean meal and VB6 were decided as factors, gloeostereum incarnatum mycelia biomass as response value,, optimized the liquid culture medium of Gloeostereum incarnatum to the nutritional content by response surface design CCD (central composite design) design. the best medium composition is:maltose 12.96 g/L, soybean meal 11.68 g/L, VB6 1.585 g/L, K2HPO4 3.5 g /L, MgSO4 2.3 g/L, pH natural. Design-expert analyzes the results obtained fermentation biomass quadratic regression model:Y= 10.50+0.75X1+0.63X2+0.20X3+0.044X1X2 0.075X1X3+0.088X2X3-0.51X12-0.79X22-0.55X32, Y is the biomass, X, addition of maltose, X2 for the addition of both soybean meal, X3 is VB6 dosage; analysis of variance was significant (p<0.01), correlation coefficients were 0.9257. This suggest that Gloeostereum incarnatum response surface optimization of fermentation medium are reasonable scale production, which can provide a theoretical basis for the application of enlargement.In the polysaccharide extraction study, L9 (34) orthogonal experimental design was used. solid to liquid ratio, extraction time and extraction temperature were selected to optimize the extraction process. The result of the best conditions for extraction is solid to liquid ratio 1:60, the extraction time 7h, extraction temperature 95 ℃. On this condition, elm spore mycelium polysaccharide ear total sugar extraction rate is 15.44%, polysaccharide content is 8.6%. For purifying polysaccharide, the polysaccharide by elm on the kind of ear volume 4mL, elution speed:5mL/8min,elution best.Using the Oxford cup to study bacteriostasis activity of Gloeostereum incarnatum polysaccharide. The results suggest that the mycelium polysaccharide inhibited the growth of Escherichia coli, Salmonella typhi, Staphylococcus aureus, Proteus, Shigella dysentery microorganisms, showed the significent effect on Bacillus subtilis and Shigellae infection.
