Corruption is one of the main factors of food waste. The common effect of inside and outside environment and microbial lead to food spoilage. A lot of type of microorganisms cause food spoilage. As the food corruption caused a huge waste. In ancient times, the way people use salt marshes to preserve food. With the development of science and technology, the methods people use salt marshes to preserve food is becoming more and more. Such as low temperature cold storage, a preservative, and high temperature sterilization. The way to corrosion protection in most Ready-to-eat food is to add preservatives. But the preservative is a kind of chemical reagents. It is harmful to human body.so, in recent years, a lot of microbiologist start trying to use microbial agents to suppress food of corruption. Microbial agents are some metabolites which some microorganism produce. It can inhibit the growth of corruption, and harmless to the human body itself. We must screen a Suitable strain in order to get these metabolites.In this paper we Isolate a strain of corruption from Corrupt food Pseudomonas gessardii. GFP,the fluorescent protein is leaded to this strain of corruption to build a engineering bacteria through three close combination.We named it PJ-9. It was as the inhibitory action. Amplification of JK-3 sequence of 16 s and construct phylogenetic tree, and determined by physiological and biochemical experiments for Weissella, It is G+.The average radius of bacteriostasis of PJ-9 is 0.623 cmPickled beans, pickled cabbage, pickles and pickled chilli chicken feet were used as the filter material.The diluents of four kinds of materials of immersion are coated to the LB and MRS culture medium. Learning the right amount of the diluents of PJ-9 bacteria night to mixed with LB medium fully then convert medium plate. The bacterium from four kinds of materials are planted on LB night tablet mixed with PJ-9 with sterilization of toothpick by using some kinds. I have screened for 16 strains ofbacteria according to whether produce the bacteriostatic circle including 5 strains screened out from the pickled beans,2 strain from pickled cabbage,6 strains from sauerkraut,3 strains from pickled chicken feet. Through further after screen, and then according to the size of the bacteriostatic circle and bacteriostasis of the length of time, I have screened for one strain of inhibitory action as research object.The fermentation conditions of Weissella sp. JK-3 were optimized by single factor. The optimal conditions were as follows:1% sucrose,1% yeast extract,0.05% dipotassium phosphate,30℃, 100ml/250 ml(100 ml in 250 ml flask),44 h, Initial pH 6.0,5% of Inoculation quantity.
