Zein can be dissolved in different concentration of ethanol and isopropyl alcohol solution and made film by coating. The purpose was studied the aggregate behavior of zein molecules dissolved in different solvents and the solvent characteristics influenced on protein film properties.In order to research on the properties of zein dissolved in different solutions, the SDS-PAGE, viscosity method, sulfhydryl method, fluorescence probe method and dynamic light scattering method were used in the paper. The result showed that zein was dissolved in solution formed alpha zein and the size of molecular weights were 22 kDa and 25 kDa. Viscosity properties of zein dissolved in solution showed zein solution belongs to pseudoplastic fluid; Zein dissolved in isopropyl alcohol solution and ethanol made more numbers disulfide bonds. The surface hydrophobicity of zein dissolved in 90% ethanol and isopropyl alcohol solution was highest. There were smaller aggregated proteins in ethanol and isopropyl alcohol solution and the aggregated proteins got bigger with the increasing of concentration of the solution obtained by Dynamic light scattering experiment.Zein film made by using different concentrations and different kinds of solution showed than zein can be made film in 75%-95% concentrations of solution.The properties of zein film made by using 75%-95% concentrations kinds of alcohol solution by measuring mechanical properties, thermal stability, water resistance and apparent morphology were studied. The results showed zein film made by using 90% concentrations of solution had better the mechanical properties, thermal stability and hygroscopicity properties. FTIR of zein film were analyzed and the result indicated that ethanol and isopropanol changed content of protein secondary structure.In order to control protein solution to make specific condensed structure for preparation of high water resistance and high mechanical food film and for preparation of porous biological cells medical organization support. The paper provided the basic theory.
